组织工程自体骨修复兔大块骨缺损

Auto-tissue engineering bone for rabbit segmental bone defects

摘要: 目的:研究组织工程自体骨的植入对大块骨缺损修复的影响。方法:培养兔的自体原代骨髓间充质干细胞,制备壳聚糖-明胶网络/羟基磷灰石多孔材料-重组人BMP2(CS-Gel/HA-rhBMP-2)复合材料。将骨髓间充质干细胞作为种子细胞与CS-Gel/HA-rhBMP-2进行复合。截去兔桡骨中段7 mm长度的骨组织,建立骨缺损动物实验模型。实验组(每只兔的右侧)植入CS-Gel/HA-rhBMP-2-种子细胞复合材料,对照组(每只兔的左侧)植入CS-Gel/HA材料,两组均不做内外固定,逐层缝合伤口。分别在术后第4周、8周和12周对缺损处进行X-线评分和组织学评分。结果:术后第4周实验组X-线和组织学评分与对照组相比无明显差别, 8周和12周实验组X-线和组织学评分均优于对照组,具有统计学意义(P<0.05)。结论:实验中所用移植修复材料具有优异的骨传导性、骨诱导性,可以缩短骨缺损的修复时间。

Abstract: Objective: Implantation of auto-tissue engineering bone to cure the rabbit segmental bone defects. Methods:Culture the rabbit mesenchymal cell, prepare the auto-tissue engineering bone with CS-Gel/HA-rhBMP-2. Bone mesenchymal stem cells as seed cells and carry out complex CS-Gel/HA-rhBMP-2.The establishment of animal models of bone defect is to amputate forelimb mid-7mm length of the rabbit bone tissue . In experimental group(right side),treat the rabbit segmental bone defect model with CS-Gel/HA-rhBMP-2-seed cell,in control group (left side),the implantation is CS-Gel/HA.Not do internal and external fixation,sew up the wound layer by layer.Evaluate the grade of X-ray and histology after treatment 4,8,12 weeks.Results:There are no significant differences of X-ray grade and histology grade between experiment group and control group at 4 weeks after treatment, but the X-ray grade and histology grade is better in experiment group than in control group at 8,12weeks after treatment,data are statistically significant (p<0.05).Conclusion: The auto-tissue engineering bone can cure the rabbit segmental bone defects.
