日本血吸虫感染小鼠CD8α-CD11c+型树突状细胞对哮喘抑制作用的肺部病理学研究

The inhibitory effect on asthma by CD8α-CD11c+ dendritic cells from Schistosoma japonicum infected mice

摘要: 摘 要:目的 通过过继转移日本血吸虫感染鼠CD8α-CD11c+树突状细胞(DC) ,研究CD8α-CD11c+DC在抑制过敏性哮喘中的作用。方法 用磁珠分选(MACS)方法纯化日本血吸虫感染鼠及正常鼠DC亚群CD8α-CD11c+细胞。将20只BALB/c小鼠随机分为4组,A组为过继转移日本血吸虫感染鼠CD8α-CD11c+ DC并诱发过敏性哮喘组,B组为过继转移正常鼠CD8α-CD11c+ DC并诱发过敏性哮喘组,C组为单纯过敏性哮喘组,D组为生理盐水对照组。A、B两组小鼠经尾静脉过继转移5 ×105 DC,1h后A、B、C3组均开始诱发哮喘。4周后处死小鼠,取小鼠肺组织进行病理学检查。 结果 与B、C 组比较,A组肺部炎症明显减轻, 按Underwood标准,A、B、C 、D 4组总评分分别为7.67±2.34、l0.86±1.35、11.75±2.22、4.5±1.73 ,差异有统计学意义( P<0.05)。结论 日本血吸虫感染鼠CD8α-CD11c+D C明显抑制过敏性哮喘的发生。

Abstract: Abstract: Objective To study the inhibitory effect on allergic asthma by adoptive transfer of    CD8α-CD11c+ dendritic cells (DC) from Schistosoma japonicum-infected mice. Methods CD8α-CD11c+DC from S.japonicum-infected mice and naive were isolated and purified using microbeads respectively,. Twenty BALB/c mice were divided into four groups randomly.5×105 CD8α-CD11c+DCs from S.japonicum-infected mice were injected into the tail vein of each mouse in Group A,while 5×105 CD8α-CD11c+ DCs of na?ve were injected into the tail vein of each mouse in Group B. One hour after injection,mice in Group A、B and C were sensitized and challenged with ovalbumin to induce asthma.Four weeks later,all the mice were killed,and the left lungs were cut for histopathologica1 analysis.Results Compared with mice in Group B and C,pulmonary inflammation of mice in Group A diminished markedly.According to Underwood’s standard,average total scores of Group A,B,C and D were 7.67±2.34,l0.86±1.35,11.75±2.22and 4.5±1.73 respectively ( P< 0.05). Conclusion CD8α-CD11c+ DC from S.japonicum infected mice can significantly inhibit allergic asthma.
