瘦素在大鼠心肌缺血-再灌注损伤中作用的探讨

Role of leptin in the mouse myocardial ischemia / reperfusion injury*

摘要: 【摘要】 目的 通过观察瘦素对心肌缺血/再灌注损伤(MIRI)大鼠模型的心肌影响,探讨瘦素在心肌缺血/再灌注损伤(MIRI)中的作用。方法24只雄性SD 大鼠随机分为四组:①假手术组(6只);②标准MIRI组(6只);③瘦素预处理组(1eptin+MIRI组)(6只);④瘦素处理组(MIRI+leptin组)(6只)。四组在MIRI手术后120min处死大鼠,观察心肌梗死面积、心肌瘦素水平以及血清瘦素、磷酸肌酸激酶(CK) 、 乳酸脱氢酶(LDH) 、 天冬氨酸转氨酶(AST)、 肌钙蛋白(TnT)水平。结果:瘦素处理增加心肌梗死面积和心肌瘦素水平及血清瘦素、磷酸肌酸激酶(CK) 、 乳酸脱氢酶(LDH) 、 天冬氨酸转氨酶(AST)、肌钙蛋白(TnT)水平。瘦素预处理可减少心肌的梗死面积和心肌瘦素水平及血清瘦素、磷酸肌酸激酶(CK) 、 乳酸脱氢酶(LDH) 、 天冬氨酸转氨酶(AST)、肌钙蛋白(TnT)水平。结论:瘦素处理和高水平的瘦素加重了心肌缺血/再灌注损伤,瘦素预处理可通过减少心肌和血清的瘦素水平以减轻了心肌缺血/再灌注损伤。

Abstract: 【Abstract】 Objective To study the effect and role of leptin on myocardial ischemia / reperfusion injury (MIRI) in mouse。Methods:Twenty four male rat were randomized into 4 group:①the sham-operation group;②the myocardial ischemia reperfusion group (MIRI group),established by blocking left coronary artery anterior descending branch for 30 min followed by reperfusion ③the leptin pretreatment group(leptin+ MIRI group),established by injecting intraperitoneally leptin at 30 min before ischemia; ④the leptin post—treatment group(MIRI +leptin group),established by injecting intraperitoneally leptin at 30 min after reperfusion。Blood and heart samples were harvested from each mouse at 120 min after reperfusion。We then analyzed the samples for leption level associated with MIRI。Results: Leptin treatment increased myocardial infarct size and myocardial leptin levels and serum leptin、creatine kinase (CK)、lactate dehydrogenase (LDH)、aspartate aminotransferase (AST)、Troponin T(TnT) levels. Leptin pretreatment reduces myocardial infarct size and myocardial leptin levels and serum leptin、creatine kinase (CK)、lactate dehydrogenase (LDH)、aspartate aminotransferase (AST) 、Troponin(TnT)levels. Conclusion: Treatment and high level of leptin increased the myocardial ischemia / reperfusion injury, leptin pretreatment reduced myocardial ischemia / reperfusion injury。
