蜀葵根预防和治疗大鼠肾草酸钙结石的效果评价及机制初探

Effects and Mechanism of Alcea Rosea Roots in Ethylene Glycol Induced Nephrolithiasis in Rats

摘要: 目的 评价蜀葵根预防及治疗大鼠肾草酸钙结石的效果,并初步探讨其机制。 方法 将60只雄性Wistar大鼠,随机分为空白对照组,模型组,蜀葵根低剂量预防组,高剂量预防组,低剂量治疗组,高剂量治疗组。空白对照组给予自由进食水,模型组给予1%乙二醇自由饮水及进食,预防组给予1%乙二醇饮水及蜀葵根提取物灌胃,治疗组开始给予1%乙二醇饮水,第15天开始添加蜀葵根提取物。实验结束时,利用大鼠尿液、血液及肾脏匀浆标本,检测大鼠尿量、钙、磷、镁、尿素、肌酐、草酸及匀浆参数。显微镜下观察肾组织切片中草酸钙结晶沉积及病理变化。 结果 与空白对照组相比,模型组大鼠尿量、血肌酐、尿素,尿草酸、尿钙、肾脏匀浆丙二醛(MDA)及肾组织内草酸钙结晶评分明显升高(P<0.05),肾脏匀浆谷胱甘肽过氧化物酶(GSH)、超氧化物歧化酶(SOD)含量明显下降(P<0.05)。与模型组比较,预防组、治疗组大鼠血肌酐、尿素,尿草酸、尿钙、肾脏匀浆MDA含量及肾组织内结晶评分明显下降(P<0.05),肾组织匀浆GSH、SOD含量明显升高(P<0.05)。肾组织病理学显示蜀葵根提取物预防组及治疗组大鼠肾内草酸钙结晶沉积较模型组明显减少,肾组织损伤较模型组明显减轻。 结论 蜀葵根可以有效预防和治疗大鼠肾草酸钙结石,保护肾功能。

Abstract: Objective To study the beneficial effect and mechanism of Alcea rosea roots in ethylene glycol induced nephrolithiasis in rats. Methods We randomly divided 60 male Wistar rats into six groups, control group, model group, Alcea rosea roots lower-dose preventive group, Alcea rosea roots high-dose preventive group, Alcea rosea roots lower-dose curative group, Alcea rosea roots high-dose curative group. Control group was provided free access to food and normal drinking water, model group received ethylene glycol drinking water and was fed with normal chow, preventive group as model group received Alcea rosea roots in low and high dose, curative group as model group received Alcea rosea roots in low and high dose from day 15 to day 28. At the end of the experiment, various renal functional and injury markers such as urine volume, calcium, phosphate, magnesium, urea, creatinine, and oxalate were evaluated using urine, serum, and kidney homogenate. The kidneys were removed and prepared for histological evaluation of calcium oxalate deposits. Results The model groups increased the urine output, urea, creatinine, 24h urine Ca2+, and oxalate and MDA compared to the control group(P<0.05). The preventive and curative groups increased the GSH and SOD compared to the model group(P<0.05). The preventive and curative groups reduced the urea, creatinine, 24h urine Ca2+, urine oxalate, MDA compared to the model group(P<0.05). It was also reduced the number and size of calcium oxalate crystal deposits and protected the kidneys from serious damage compared to the model group. Conclusion The extract of Alcea rosea roots can prevention and treatment calcium oxalate urinary stone formation in rats and protect renal function.
