微生物培养法测定血清中的叶酸浓度

摘要: 摘要 目的:建立一种准确、简便、廉价、高效且易于推广的血清叶酸微生物培养测定方法。方法:利用干酪乳酸菌生长需要叶酸作为营养的特点,使用活性炭粉末吸附试验用水中的有机杂质,建立血清叶酸浓度测定方法。结果:该方法测定范围0~0.6μg/L;检出限0.017μg/L;测定高、中、低三种浓度血清样品,批内、批间变异系数均小于5%,回收率为98.1%~102.4%;与化学发光方法比较,相关系数0.963,但二者存在一定差异;试剂在-20℃冻存可稳定6个月以上。结论:该法测定血清叶酸结果准确,且简便、廉价、高效、易于推广,适用于大样本筛查。

Abstract: Abstract Objective: To establish an accurate, simple, cheap, efficient and promotable biological assay for serum folate. Methods: Based on the property of folate for growth of lactobacillus, serum folate test method was established by use of acticarbon powder adsorption of organic impurities in the test water. Results: The measure range was 0 ~ 0.6μg / L and the detection limit was 0.017μg / L. The intra and inter CVs were less than 5% and the recovery rate was 98.1 % ~102.4%. The correlation coefficient was 0.963 comparing with chemiluminescence method. The reagent could be stable more than 6 months when frozen at -20 ℃. Conclusion: This serum folate test method was accurate, simple, cheap, efficient,promotable, and applicable to the screening for the large samples.
