1.0%与0.2%葡萄糖环境中口腔变形链球菌基因表达的差异*

摘要: [摘要] 目的:观察葡萄糖营养条件对变形链球菌基因表达的影响。方法:变形链球菌分别在1.0%、0.2%葡萄糖条件下培养,利用前期构建的变形链球菌基因表达谱检测芯片筛选两种培养条件下变形链球菌中差异表达的基因,以观察环境葡萄糖供给情况对变形链球菌基因表达的影响。结果:相对于0.2%葡萄糖,在1.0%葡萄糖条件下变形链球菌中有约57种基因表达发生显著变化,其中与变形链球菌糖转运系统和能量代谢、粘附等细菌毒力、DNA复制、重组、修复、信号传递与调控系统、细菌表面成分等相关的41种基因表达明显上调,而一些参与细胞生理过程的感受态蛋白和酶的16种基因表达显著下调(P

Abstract:

[Abstract] Objective  To detect the differently expressed genes of Streptococcus mutans (S. mutans ) cultured in 1.0% glucose and 0.2% glucose. Methods  S. mutans was cultured in 1.0% and 0.2% glucose. The genetic expression of S. mutans under different conditions was detected using gene chips built previously. Results  In 1.0% glucose environment, expressions of 57 S. mutans genes significantly changed compared with 0.2% glucose. Forty-one genes associated with sugar transport and energy metabolism, virulence, pathogenesis, DNA metabolism and regulatory functions were significantly up-regulated. While 16 genes encoding some competence proteins and enzymes related to cellular processes were significantly down-regulated. Conclusion  The environmental glucose affects multiple gene expressions of S. mutans.
