p38抑制剂对辐射损伤小鼠中免疫细胞的作用

The effects of p38 inhibitor on the immune cells in irradiated mice

摘要: 目的 探讨p38抑制剂SB203580(SB)对免疫细胞辐射损伤的作用。方法 实验设对照组、照射组、SB组。以6.0Gy 137Cs γ射线全身照射SB组和照射组的C57BL/6小鼠。照射24h后开始腹腔注射SB(15mg/kg),隔日1次,共给药5次,末次给药24h后处死小鼠,取外周血进行测定,并检测CD4、CD8、B220表达,取骨髓细胞测定ROS水平。结果 与对照组相比,照射组外周血白细胞(WBC)、CD8、B220细胞比例明显下降,骨髓ROS水平升高(P<0.05)。与照射组比较,SB组外周血白细胞增高,CD8比例上升,骨髓ROS水平下降(P<0.05)结论 p38抑制剂(SB)对小鼠6Gy照射后造血免疫损伤有一定的保护作用,其机制可能与其降低照射后细胞内ROS水平有关。

Abstract: Objective To observe the effects of p38 inhibitor SB203580 (SB) on the immune cells in the radiated mice. Methods C57BL / 6 mice were divided into control group, irradiated group, SB group. The control group received sham irradiation, and the rest accepted 6 Gy total body irradiation. Mice in SB group were intraperitoneally injected 15mg/kg at 1,3,5,7,9 day after irradiation. After mice exposed to 6Gy irradiation 10 day, the peripheral bloods and bone marrow cells were collected. The immune cells phenotypes in peripheral bloods were analyzed by flow cytometry, ROS (reactive oxygen species) level of bone marrow cells were detected by kits. Results Compared to control group,the proportion of CD8, B220 and WBC in the irradiated mice significantly decreased, ROS level of bone marrow cells increased (p <0.01). Compared to irradiation group, the CD8 and WBC in SB treated group increased, ROS level in bone marrow cells of SB treated group decreased (p <0.05). Conclusion The results indicate that p38 inhibitor can protect the immune cells from radiation injury by lowing the ROS level and it might used as a therapeutic method for the radiation injury .
