nPKC和cPKC调节骨骼肌葡萄糖转运体4型转位的研究

nPKC and cPKC Modulate Translocation of Glucose Transporter 4 in Skeletal Muscle Cells

摘要: 摘要 目的:探讨新颖型蛋白激酶C(nPKC)和传统型蛋白激酶C(cPKC)调节骨骼肌葡萄糖转运子4(GLUT4)转位的作用。方法:测定C2C12GLUT4myc肌管经PKC激活剂佛波酯(PMA)和PKC抑制剂Go6976,Go6983处理后,细胞表面的GLUT4myc水平。结果:PMA急性刺激可以增加细胞GLUT4myc转位,Go6976和Go6983不同程度抑制PMA刺激的GLUT4myc转位,PMA慢性孵育,部分抑制PMA急性刺激的GLUT4myc转位。结论:激活nPKC或cPKC增强骨骼肌葡萄糖转运子4转位。

Abstract: nPKC and cPKC Modulate Translocation of Glucose Transporter 4 in Skeletal Muscle Cells LI Jinru,NIU Wenyan,CHU Guilan Basic Mdical College, Tianjin Medical University,Tianjin 300070, China Abstract Objective: To evaluate the effects of novel protein kinase C (nPKC) and conventional protein kinase C(cPKC) on modulation of translocation of glucose transporter 4 (GLUT4) in skeletal muscle cells. Methods: To determinate the level of GLUT4myc translocation on cellular surface by treatment with PKC activator PMA and PKC inhibitors Go6976, Go6983 in C2C12GLUT4myc myotubes. Results: Acute stimulation of PMA can significantly increase the translocation of GLUT4myc, Go6976, Go6983 inhibited the translocation of GLUT4myc in different degrees. Chronic stimulation of PMA partly inhibited the translocation of GLUT4myc on cellular surface. Conclusion: Activation of nPKC or cPKC enhance translocation of GLUT4 in Skeletal muscle cells.
