H102对APP转基因小鼠突触相关蛋白表达的影响

Effects of H102 on related protein of synaptophysin of APP trangenic mouse

摘要: 摘要 目的 观察H102对APP转基因小鼠脑内shank1,PSD-95,突触素表达的影响。 方法 将APP转基因小鼠(n=16)随机分为模型组和给药组,每组8只。并设同月龄同背景C57BL/6J小鼠为正常对照组。给药组侧脑室注射H102生理盐水溶液,对照组和模型组侧脑室注射等体积生理盐水,3μl/d, 共30天后行行为学检测即水迷宫测试,然后固定脑组织,利用免疫组化及westen blot方法测定小鼠脑组织突触素、PSD-95及Shank1表达。结果 [1]模型组皮层和海马出现突触素、PSD-95 和 Shank1表达减少;注射H102后突触素,PSD-95 和 Shank1表达增多,接近正常对照组;⑵ 定位航行实验H102给药后APP转基因小鼠逃避潜伏期较给药前显著缩短(P<0.01);跨越平台次数较给药前显著增加(P<0.01);第三象限停留时间显著增加(P<0.01)。结论 侧脑室注射H102可以提高突触相关蛋白突触素、PSD-95和Shank1的表达,改善突触的可塑性;修复神经元、突触的形态学损伤;提高APP转基因AD模型小鼠空间学习能力。H102是一种很有发展前景的AD治疗药物。

Abstract: [ABSTRACT] AIM To observe the effects of H102 on related protein of synaptophysin of APP trangenic mouse. METHODS The APP transgenic mice were radomly divided into three groups which are modle group ,H102 treatment group, and the control group which of C57BL/6J mice with the same age and background . Modle geoup and normal control group were treated with saline solution while the H102 treatment group was treated with compounds (80μmol/L H102 in saline solution ),med icine or saline was infused in lateral cerebral ventricle every day.after flished that do water maze testing Immunohistichemical stain and westen blot were done with synapse,PSD-95, shank1.RESULTS [1]Morris water maze test showed that the mean escape latent period(MELP) ,the time of the mice across the flat, and the time of traversing through flat roof,and the time which the mice stay in the third space,were was obviously shorten after H102 treatment compare with the modle group(P<0.01),and similer to the nomal group(P>0.05).[2]The content of synapse,PSD-95 and shank1 were highter in H102 treatment group than in modle group (P<0.01)and simile to the nomal group.CONCLUSION H102 can improve the APP trangenic mouse’ ability of remember obviously,and the content of synapse,PSD-95 and shank1 in APP trangenic mouse’ briarn become higher.H102 is a greate and effective durg which will be treated with AD.
