RGS4 蛋白在小儿肾母细胞瘤组织中的表达及临床意义

Clinical significance of expression of RGS4 in pediatric nephroblastoma tissue

摘要: 摘要: 目的 分析 G 蛋白信号调节蛋白 4 (RGS4) 在小儿肾母细胞瘤组织和癌旁组织中的表达差异, 探讨 RGS4 与小儿肾母细胞瘤发生发展的关系。方法 收集 37 例小儿肾母细胞瘤患者术后肾癌组织及 8 例癌旁组织, 采用免 疫组织化学法检测 RGS4 蛋白的表达水平; 另收集患者手术切除新鲜癌组织及对应癌旁组织标本各 8 例, 采用 qRTPCR 和 Western blot 分别检测 RGS4 mRNA 和蛋白的表达水平。结果 免疫组织化学染色结果显示, RGS4 蛋白在 小儿肾母细胞瘤组织及癌旁组织中均存在阳性表达, 其在小儿肾母细胞瘤组织中的高表达率低于癌旁组织 [37.83% (14/37)vs. 87.5%(7/8), χ2=4.675, P < 0.05]; RGS4 mRNA 在小儿肾母细胞瘤组织中的表达水平显著低于癌旁组织 (1.064±0.549 vs. 5.374±0.735, t=13.290, n=8, P < 0.01); Western blot 结果显示, RGS4 蛋白在小儿肾母细胞瘤组织中 的表达量低于癌旁组织 (0.301±0.092 vs. 0.779±0.041, t=13.424, n=8, P < 0.01)。结论 RGS4 在小儿肾母细胞瘤组织 中表达下调, 可能与小儿肾癌的发生发展有关。

Abstract: Abstract: Objective To investigate the expression levels of regulator of G-protein signaling 4 (RGS4) in pediatric nephroblastoma and pericancerous tissues, and explore the relationship between RGS4 and the occurrence and development of pediatric nephroblastoma. Methods Thirty- seven samples of pediatric nephroblastoma tissues and 8 samples of pericancerous tissues were collected after surgery to detect the expression of RGS4 protein by immunohistochemistry. Another 8 samples of fresh cancer tissues and corresponding pericancerous tissues were collected to detect the mRNA and protein levels of RGS4 by qRT-PCR and Western blot assay, respectively. Results Immunohistochemistry results showed that RGS4 protein was positively expressed both in pediatric nephroblastoma and pericancerous tissues, and its high expression rate was lower in pediatric nephroblastoma than that in pericancerous tissues [(37.83% (14/37) vs. 87.5% (7/8), 2χ =4.675, P<0.05]. The expression level of RGS4 mRNA was significantly lower in pediatric nephroblastoma than that in pericancerous tissues (1.064 ± 0.549 vs. 5.374 ± 0.735, t=13.290, n=8, P < 0.01). Western blot results showed that the expression level of RGS4 protein was lower in pediatric nephroblastoma than that of pericancerous tissues (0.301±0.092 vs. 0.779 ± 0.041, t=13.424, n=8, P < 0.01). Conclusion The expression level of RGS4 is down- regulated in pediatric nephroblastoma, which may be related to the occurrence and development of pediatric nephroblastoma.
