甜瓜蒂 3 种提取物体外抗肿瘤活性的比较研究

Comparative study of in vitro anti-tumor activities of three different extracts of Pedicellus Melo

摘要: 目的 观察甜瓜蒂 3 种提取物对体外培养肿瘤细胞增殖的影响, 并初步探讨其可能的作用机制。方法以溶剂提取法分别得到甜瓜蒂的水提取物、 75%乙醇提取物、 95%乙醇提取物。采用四甲基偶氮唑蓝(MTT)法检测这 3 种提取物对人胃腺癌细胞 SGC-7901、 人肺癌细胞 NCI-H460、 人肝癌细胞 SMMC-7721、 人白血病细胞 K562 及小鼠成纤维细胞 L929 的增殖抑制作用。应用流式细胞术检测样品对细胞周期和凋亡的影响。结果 甜瓜蒂水提取物、 75%乙醇提取物、 95%乙醇提取物作用 72 h 后, 在测定的浓度范围内对 SGC-7901、 NCI-H460、 SMMC-7721、 K562 及 L929 等 5 种细胞均表现出不同程度的生长抑制作用, 量效关系基本明确。甜瓜蒂 75%乙醇提取物、 95%乙醇提取物组对各细胞的半数抑制浓度(IC50)均低于水提取物组(均 P<0.05), 其中 95%乙醇提取物组对 SGC-7901、 SMMC-7721、 K562 及 L929 细胞的 IC50明显低于 75%乙醇提取物组(均 P<0.05)。5 种细胞中 SGC-7901 对甜瓜蒂提取物较为敏感, 该细胞经 0.4、 0.6、 0.8 mg/L 的 95%乙醇提取物作用 48 h 后, 细胞凋亡率分别为(8.32±0.55) %、(11.06±1.15) %、(12.25±1.51) %; 细胞被阻滞在 S 期、 G2/M 期。结论 甜瓜蒂 95%乙醇提取物体外对 SGC-7901 具有显著的抑制作用, 其机制可能与诱导细胞凋亡和抑制细胞周期有关。

Abstract: Objective To study the antitumor effects of three different extracts from Pedicellus Melo on the proliferation of cancer cells in vitro, and discuss the possible mechanism. Methods Pedicellus Melo was extracted by water, 75% ethanol and 95% ethanol, respectively. The anti-cancer activities in vitro were detected by MTT assay on five tumor cell lines SGC- 7901, NCI-H460, SMMC-7721, K562 and L929 cells. Flow cytometry was used to detect the cell cycle and apoptosis of different cell groups. Results After 72-hour treatment with water, 75% ethanol and 95% ethanol extracts, inhibitory effects were found in five tumor cell lines, which showed dose-dependent manner on the proliferation of the cells. The IC50 values were significantly lower in the two ethanol extracts than those of water extract (P<0.05) . And the IC50 values of 95% ethanol extract on SGC-7901, SMMC-7721, K562 and L929 cells were significantly lower than those of 75% ethanol extract (P <0.05). All the extracts were sensitive to SGC- 7901 cells. Apoptotic rates of SGC- 7901 cells treated with 95% ethanol extract of the concentrations of 0.4, 0.6 and 0.8 mg/L for 48 hours were (8.32±0.55)%, (11.06± 1.15)% and (12.25±1.51)%. The cell cycle was arrested in S and G2/M phases. Conclusion Results demonstrate that 95% ethanol extract of Pedicellus Melo has significant inhibitory effect on SGC-7901 cells in vitro, which may be related to the induced apoptosis and the inhibition of cell division.
