小鼠原位气管移植模型手术技巧及改良

摘要:目的 探讨改良原位气管移植模型的手术方法以提高生存率, 减少手术时间。方法 清洁级 C57BL/6 (供
体) 和 BALB/c (受体) 小鼠各 30 只, 依体质量相近配对并随机分为 3 组。采用 “经口插管气道内支架” 吻合, 原位气管
移植建立动物模型, 并记录手术时间。术后观察实验动物生存期及一般状况, 并分别于术后 1530 60 d 各处死
10 只, 取出气管, HE 染色观察移植气管的组织病理变化, 计算气管阻塞率。结果 受体小鼠均存活至取气管时, 手
术时间: 供体气管摘除(7.0±1.0) min, 受体气管摘除(7.0±1.0) min, 经口插入套管(1.0±0.5min, 供体气管吻合(20.0±
4.0min, 手术总时间 (43.0±10.0min。移植后气管淋巴细胞及中性粒细胞浸润, 气管阻塞率于 1530 60 d 逐渐增
加, 符合闭塞性细支气管炎病理特点。结论 经口插管气道内支架吻合法建立小鼠原位气管移植模型操作简便, 手
术技巧和设备要求低, 手术成功率高。不需要专业的外科技巧及手术设备。

Abstract:

Abstract: Objective To explore a simple and effective way of establishing a mice orthotopic tracheal transplantation
model in studying bronchiolitis obliterans to reduce operation time. Methods Tracheas from C57BL/6 mice (n=30) were im⁃
planted into Balb/c mice (n=30). Grafts and hosts were matched according to body weight. "Orotracheal intubation and endostent" was used for trachea anastomose and animal model was establish. Operating time was also noted. Grafts were harvest⁃
ed on Days 15, 30 and 60 after transplantation, using HE staining to observe pathological changes of allograft. Trachea block⁃
ing rate was also calculated. Results Until survival of trachea transplantation, the operative time of donor′s and the recipi⁃
ent′tracheas resection were (7.0±1.0) min and (7.0±1.0) min respectively. "Orotracheal intubation the endo-stent" establish⁃
ment cost1±0.5min. Trachea anastomose took20.0±4.0min and the operation time last43.0±10.0min. Conclusion
The improvement of orthotopic tracheas transplantation using "Orotracheal intubation the endo-sten" for trachea anastomose
is esay-operated and did required complicate skills and instruments with high successful rate. So it is an ideal and stable
model in studying bronchiolitis obliterans.
