胎盘TLR3分布及与HBV宫内感染的关系初探

Relationship between TLR3 expression in placenta and intrauterine infection of hepatitis B virus

摘要: 目的:了解Toll样受体(TLR)3在外周血HBsAg阳性母亲胎盘组织中的表达及分布,探讨其与HBV宫内感染的关系。方法:采用SABC免疫组织化学方法结合显微镜观察胎盘组织HBsAg和TLR3的表达和分布,以胎盘各层细胞出现棕黄色颗粒判断为阳性;Abbott化学发光法检测新生儿外周血HBsAg;ELISA法检测母亲外周血HBsAg和HBeAg。结果:24例母亲外周血HBsAg均阳性,除2例因胎盘组织脱落不计外,22例中17例胎盘组织中HBsAg呈阳性,6例新生儿发生HBV宫内感染,宫内感染率25%(6/24),胎盘HBV感染率77.27%(17/22)。HBsAg分布于蜕膜细胞、滋养层细胞、绒毛间质细胞、绒毛毛细血管内皮细胞。17例HBsAg阳性的胎盘组织中16例TLR3阳性,阳性细胞较少,主要在滋养层细胞和绒毛间质细胞。1例宫内感染的新生儿、HBsAg阴性的胎盘组织5例中的4例、外周血HBsAg阴性的6例正常胎盘组织切片上及空白对照均未检测到TLR3阳性表达。结论:外周血中HBV对TLR3可能仅有微弱的激活作用。

Abstract: Abstract Objective:To study on the distribution of toll-like receptor 3 (TLR3) in human placenta and their role in the HBV intrauterine infection. Methods:Twenty two full-term placental tissues of HBsAg carriers and 6 ones of HBsAg sero-negative pregnant women were collected. The HBsAg and TLR3 in the placental tissues were investigated by StreptAnidin-Biotin peroxidase Complex (SABC) immunohistochemical staining. Neonatal HBsAg and HBeAg was tested by Chemiluminescence. HBsAg in maternal sera were detected by ELISA. Results:The HBsAg positive rate in the full-time placental tissues from HBsAg carrier pregnant women was 77.27% (17/22). The positive signals of HBsAg could locate in the cytoplasms of decidual cells, trophoblastic cells, villous meseachymal cells and villous capillary endothelial cells. The positive signals of TLR3 major locate in the cytoplasm of trophoblastic cells and villous meseachymal cells in the HBsAg-positive placental tissues. TLR3 could not be found in the placenta of HBsAg sero-negative pregnant women. Conclusion: Preliminary considering, there may be only weak response of TLR3 to HBV stimulation.
