天津地区风疹病毒的分子特征分析

Molecular Characterization of Rubella Viruses Isolated in Tianjin

摘要: 摘要 目的:了解2009年天津地区风疹病毒(Rubella Virus,RV)的分子特征。方法:采用Vero/SLAM细胞培养法分离出10株风疹病毒,用RT-PCR方法扩增10株风疹病毒E1基因1170个核苷酸片段,并对该PCR产物进行序列测定和分析。分离株与WHO各基因型参考株基于E1基因739个核苷酸构建基因亲缘性关系树。结果:10株风疹病毒与1E基因型代表株处于同一分支,属于1E基因型。核苷酸同源性为97.3%-100%,重要抗原位点未发生变异。血凝抑制和中和位点中,只有Tianjing09-06株在E1蛋白第234位氨基酸发生了变化(Trp234→Ser234)。同时所有天津分离的风疹毒株在E1蛋白的第338位氨基酸发生相同突变(Leu338→Phe338),该氨基酸(Phe338)可能是我国1E基因型风疹病毒所特有。结论:天津地区流行的风疹病毒为E1基因型,在基因亲缘性关系树中形成紧密相连的三簇,处于不同的进化链中,显示天津地区存在不同风疹病毒株的传播链。

Abstract: Abstract Objective: To understand the genetic characteristics of rubella viruses in Tianjin in 2009.Methods: Rubella viruses were isolated by Vero/SLAM cells and identified through RT-PCR.1170 nucleotides of E1 gene were amplified and sequenced.The phylogenetic tree of 10 rubella strains compared to the WHO reference strains for each genotype based on the 739 nucleotides of E1 gene was performed.Results: The 10 strains detected from Tianjin formed a distinct cluster in genotype 1E in the phylogenetic tree.The nucleotide sequence identities are 97.3%-100%. There was no change in the important antigenic epitope sites. Only one amino acide change(Trp234→Ser234)in Tianjing09-06 at the hemagglutination inhibition and neut ralization epitopes.Meanwhile the 10 strains isolated from Tianjin had the same amino acide change(Leu338→Phe338)in E1 protein.The result sustained other doctor’s research.The amino acide (Phe338)may only existed in Chinese 1E genotype rubella viruses.Conclusion: The rubella virus genotype circulated in Tianjin is 1E.The 10 specimens in this study clustered three close together in our phylogenetic analysis,suggesting that there may be multi-transmission routes in Tianjin.
