阿片生长因子抑制乳腺癌细胞生长的研究

摘要: 摘要 目的:探讨阿片生长因子(OGF)对MCF-7人乳腺癌细胞生长的影响。方法:通过细胞计数、MTT实验、FCM细胞周期分析观察OGF对MCF-7细胞生长的影响。结果:细胞计数显示,OGF处理组细胞数明显低于OGFr拮抗剂naloxone处理组及空白对照组(P<0.01),而后两组细胞数差异无显著性(P>0.05);MTT实验显示,OGF处理组细胞增殖抑制率明显高于naloxone处理组、OGF联合naloxone处理组及空白对照组(P<0.01),而后两组差异无显著性(P>0.05);OGF处理组MCF-7细胞G0/G1期分数明显高于naloxone处理组及空白对照组(P<0.01)。结论:OGF能抑制MCF-7人乳腺癌细胞的生长。OGFr拮抗剂能阻断这种作用。OGF阻断细胞周期于G0/G1期。

Abstract: Abstract Objective:To investigate the growth inhibitory effect of OGF and the opioid antagonist naloxone on MCF-7 human breast carcinoma cells. Methods:Cell counting、MTT essays and cell cycle distribution analysis by flow cytometry were used to detect the inhibiting growth effect of OGF on MCF- 7 cells. Results: Cell counting showed that the cell concentration reduced markedly in OGF group than other groups(P<0.01). No difference was found between naloxone and control groups(P>0.05). MTT essays indicated that the inhibiting rate of cell growth of OGF group was significantly higher than that in naloxone and OGF/naloxone groups (P<0.01). There was no significant difference among naloxone, OGF/ naloxone and control groups(P>0.05). Flow cytometry results showed that PI was lower in OGF group than in other groups(P<0.01), and no significant difference was found between naloxone and control groups(P>0.05).Conclusions: OGF could inhibit the growth of MCF-7 cells in vitro and the growth inhibitory effect of OGF can be blocked by naloxone. The cell cycle was obstructed by OGF in G0/G1 phase.
