基质金属蛋白酶组织抑制因子-1、2在子宫颈鳞癌中的表达及意义

Significance of TIMP-1 and TIMP-2 expression in uterine cervical squamous cell carcinoma

摘要: [摘要] 目的:探讨基质金属蛋白酶组织抑制因子TIMP-1和TIMP-2在宫颈鳞癌组织中的表达及其与宫颈癌临床病理特征的关系。方法:采用免疫组织化学MaxvisionTM 法,检测46例宫颈鳞癌,34例宫颈原位癌(CINⅢ)和20例正常宫颈上皮(NCE)中TIMP-1、TIMP-2的表达,分析其表达与宫颈鳞癌的临床分期、病理类型及有无淋巴结转移的关系。结果:TIMP-1在宫颈鳞癌、原位癌和正常宫颈上皮中的阳性表达率分别为54.3%(25/46)、73.5%(25/34)和20%(4/20)。TIMP-2为60.9%(28/46)、50.0%(17/34)和20%(4/20)。TIMP-1和TIMP-2蛋白阳性表达率在鳞癌组及原位癌组明显高于对照组,差异有统计学意义(P<0.01);在不同的临床分期和病理类型患者中,TIMP-1和TIMP-2的阳性表达率差异无统计学意义(P>0.05);TIMP-2在淋巴结转移阴性组的阳性表达率较阳性组高(P<0.05),而TIMP-1未发现此现象。结论:TIMP-1和TIMP-2在子宫颈鳞癌浸润和转移中起重要的作用。

Abstract: [Abstract] Objective: To investigate the expression of tissue inhibitor of metalloproteinase-1 (TIMP-1) and TIMP-2 in uterine cervical squamous cell carcinoma and the relationship between these levels and the clinicopathologic characteristics of cervical cercinoma. Methods: The expression of TIMP-1 and TIMP-2 were detected by immunohistochemical MaxvisionTM methods in 46 cases of cervical squamous cancer, 34 cases of carcinoma in situ of uterine cervix (CINⅢ), and 20 cases of normal cervical epithelium (NCE). The correlation between the expression TIMP-1, TIMP-2 and clinical stages, pathological grades, lymph node metastases in cervical squamous cancer were analyzed. Results: The expression rate of TIMP-1 in uterine cervical squamous cell carcinoma, carcinoma in situ of uterine cervix and normal cervical epithelium were 54.3%(25/46), 73.5%(25/34), 20%(4/20), respectively, and that of TIMP-2 were 60.9%(28/46), 50.0%(17/34), 20%(4/20). The positive rate of TIMP-1 and TIMP-2 in the cervical carcinomas group and CINⅢ group were obviously higher than that in NCE group(P<0.01). The protein expression of TIMP-1 and TIMP-2 did not correlate with tumor stage or histological grade(P>0.05). The positive rate of TIMP-2 in lymph node metastasis was higher than that in without lymph node metastasis(P<0.01), but there was no significant difference for TIMP-1(P>0.05). Conclusion: TIMP-1 and TIMP-2 may play a key role in invasion and lymph-node metastasis of in uterine cervical squamous cell carcinoma.
