咖啡酸苯乙酯对糖尿病大鼠视网膜VEGF和ICAM-1表达的影响

The effects of caffeic acid phenethyl ester on the expression of VEGF and ICAM-1 in the retinae of diabetic rats

摘要: 【摘要】 目的 观察咖啡酸苯乙酯对糖尿病大鼠视网膜血管内皮生长因子(VEGF )和细胞间黏附因子-1(ICAM-1)蛋白表达的影响。方法 将30只STZ诱导的糖尿病大鼠随机分为3组,糖尿病组(DM,n=10),CAPE低剂量治疗组(CAPE5,n=10),CAPE高剂量治疗组(CAPE10,n=10)。另取10只大鼠作为正常对照组(CON,n=10)。饲养三个月后,取大鼠双眼视网膜做组织匀浆,Elisa法检测视网膜VEGF 和ICAM-1的表达。结果 腹腔注射咖啡酸苯乙酯(CAPE)后,糖尿病大鼠视网膜VEGF和ICAM-1蛋白水平表达降低,低剂量治疗组可分别减少40%VEGF和29% ICAM-1蛋白的表达,高剂量治疗组可分别减少42%VEGF和44% ICAM-1蛋白的表达。 结论 咖啡酸苯乙酯可减少糖尿病大鼠视网膜VEGF 和ICAM-1蛋白的表达。

Abstract: [Abstract] Objective To observe the effccts of caffeic acid phenethyl ester on the expression of VEGF and ICAM-1 in the retinae of diabetic rats. Method Thirty diabetic SD rats Inducided by streptozotocin were divided into three groups, diabetic group (DM, n=10) ,low dose group of caffeic acid phenethyl ester(CAPE5,n=10). High dose group of caffeic acid phenethyl ester(CAPE10,n=10).Ten else rats were in normal control group.All of the rats were executed 3 months later , both side of the retinae were homogenized in each rat, and the protein level of VEGF and ICAM-1 was detected by Elisa. Results After peritoneal injection of caffeic acid phenethyl ester ,the protein level of VEGF and ICAM-1 were decreased ,in the low dose group, the protein level of VEGF and ICAM-1 were decreased by 40% and 29%,when the protein level of VEGF and ICAM-1 were decreased by 42% and 44% in high dose group. Conclusion caffeic acid phenethyl ester can reduce the expression of VEGF and ICAM-1 in retinae of diabetic rats.
