车前子多糖抗炎作用机制的实验研究

摘要: 目的:观察车前子多糖(PSP)的抗炎作用 方法:采用二甲苯致小鼠耳廓肿胀、腹腔注射醋酸所致小鼠腹腔毛细血管通透性增高实验,观察PSP的早期抗炎作用;然后,通过制作大鼠气囊滑膜炎模型, 测定渗出液容积, 渗出液中白细胞(WBC)计数、超氧化物歧化酶(SOD)、丙二醛(MDA)和肿瘤坏死因子-alpha(TNF-a)含量及血清中 SOD与 MDA含量。观察PSP对炎症中期的抗炎作用及对上述各指标的影响。同时通过小鼠棉球肉芽肿实验,观察PSP对晚期炎症的抗炎作用。结果:车前子多糖对二甲苯致小鼠耳廓肿胀、醋酸致小鼠毛细血管通透性的增加,以及小鼠棉球肉芽肿的形成均有明显的抑制作用,还能明显减少渗出液容积, 降低灌洗液中 WBC、 MDA 、TNF-a含量及血清中 MDA水平,并能提高灌洗液和血清中SOD的活性。 结论:PSP对多种炎症模型均有明显的对抗作用,其抗炎机制与抑制炎性因子的渗出、消除自由基和抑制脂质过氧化有关。

Abstract: Objective To investigate the anti-inflammatory effect of the plantain seed polysaccharides (PSP). Methods Various inflammatory models, including the swelling of ear induced by xylene in mice, the permeability of blood capillary by acetic acid in mice, viewing earlier period Anti-inflammatory Effect of PSP. Then, air sac models of synovitis in mice were set up, the amount of white blood cell (WBC), the volume and the contene of tumor necrosis factor (TNF-a) were measured in inflammatory exudan, the activity of superoxide dismutase (SOD), the contene of malondialdehyde (MDA) were measured in inflammatory exudan and blood serum to observe intermediate stage anti-inflammatory effect of PSP and the influence on above-mentioned index. Meanwhile, through the cotton-pellet granuloma of the mice, viewing advanced stage anti-inflammatory effect of PSP. Results The PSP had significant inhibitory action on auris swell induced by xylene in mice, penetration of capillary vessel in mice and the proliferation of granuloma caused by implantation cotton-pellet in mice. The amount of WBC 、the volume 、the contene of tumor necrosis factor(TNF-a) in inflammatory exudan, and the content of MDA in inflammatory exudan and blood serum were obviously lower than those in the control groups. The activity of SOD was obviously higher. Conclusion The PSP has significant anti-inflammation, which is relevant to the inhibition of the effusion on the inflammatory factor, the elimination of the free radicle and inhibition of the lipid peroxidation.
