骨髓内皮祖细胞在假孕大鼠黄体向血管内皮细胞分化的实验研究*

Experimental Study of Endothelial Progenitor Cell’s Differentiation to Endothelial Cell on Corpus Luteum of Pseudocyesis Rat

摘要: 摘要:目的:观察骨髓内皮祖细胞在假孕大鼠黄体向血管内皮细胞分化,参与血管发生;方法:雌性大鼠经137Cs全身照射后,从尾静脉移植已标记DAPI的雄性大鼠骨髓内皮祖细胞,受鼠造血重建后,制备假孕模型,于假孕后第七天应用荧光显微镜、原位杂交技术观察受鼠骨髓、外周血及黄体微血管内皮祖细胞的分布及分化情况。结果:受鼠骨髓、外周血及黄体中均有DAPI阳性细胞分布,黄体微血管可见SRY阳性内皮细胞。结论:骨髓内皮祖细胞在假孕大鼠黄体中向血管内皮细胞分化,参与黄体血管发生。

Abstract: Abstract:Objective To observe bone marrow endothelial progenitor cell’s (EPC) differentiation to endothelial cell on corpus luteum of pseudocyesis rat and the participation in vasculogenesis;Methods From caudal vein, female rats that rediated by 137Cs were transplantated EPCs marked DAPI of male rats, made pseudocyesis model of transplantated female rats after hematopoietic reconstitution,then on the 7th day, observed EPCs’ distribution and differentiation on the female rats’ bone marrow, peripheral blood and corpus luteum by fluorescence microscopy and in situ hybridization SRY;Results cells marked DAPI dispersed on models’ bone marrow, peripheral blood and corpus luteum, and male rat’s EPCs marked SRY appeared on the capillary of model rats’ corpus luteum; Conclusions EPCs start differentiation to endothelial cells and participate in vasculogenesis on pseudocyesis rats corpus luteum.
