放射性125I粒子植入对大鼠背根神经节及周围组织形态学影响

The changes of histomorphology of dorsal root ganglion and tissues around after implanting the radioactivity of 125I seeds around the interverterbal foraman of lumbar.

摘要: 摘要 目的:将不同剂量放射性碘125粒子植入大鼠腰部椎间孔附近后,观察其对背根神经节(Dorsal root ganglion, DRG)及周围组织形态学的影响。方法:选取成年雄性SD大鼠24只,体重220~250g,随机分为A、B、C、D组,每组6只。将不同剂量的放射性125I粒子植入各组大鼠左侧L5DRG周围。每组均植入2粒活度相等的125I粒子。各组放射性125I粒子的活度分别为0mCi、0.4 mCi、0.6 mCi、0.8 mCi。A组为对照组,植入的是空白粒子。于粒子植入后60天获取各组大鼠L5DRG在光镜下行组织形态学观察及计算阳性细胞率。结果 1.光镜下DRG变化:随着放射性粒子125I剂量的增加和植入时间延长,DRG细胞损伤程度从出现细胞轻度水肿,肿胀,尼氏小体减少到细胞肿胀明显,细胞核偏向一侧,核固缩、核碎裂现象增加。在0.8 mCi组粒子植入60天时损伤严重。可见大量细胞核消失、核溶解、胞浆皱缩等细胞变性病理表现。阳性细胞率随放射活度增大、时间延长逐渐增大,0.8 mCi组60天时的阳性细胞率达到73.80±2.38%。 结论 放射性125I粒子植入后可对DRG细胞造成损伤。放射性粒子的活度越高,细胞损伤的程度越严重。

Abstract: 【Abstract】Objective: To observe the effects of different radioactivity of 125I seeds on the histomorphology of dorsal root ganglion and tissues around by implanting radioactive seeds 125I around dorsal root ganglion.Methods: The experiment were carried out using twenty four adult male Sprague-Dawley rats (220-250g), randomly assigned to four groups. the different radioactivity 125I seeds were implanted around dorsal root ganglion ,respectively named 0mCi group (n=6), 0.4mCi group(n=6), 0.6 mCi group(n=6), 0.8 mCi group(n=6). CT scan were used to check the seeds whether locating in 1cm range around dorsal root ganglion.if not in the range ,the rat must be rejected. the two rats were killed at 60 days randomly to observe the histomorphology of L5 dorsal root ganglion and the surrounding tissues. Results: the implantion of 125I seeds was successful around L5 DRG in 24 rats.(2 rats were rejected, then supplement in time), CT scan ensure exact position. The cellular swelling of the 0mCi group was moderate The cell edema was more severe , the nucleus was not in center and Karyopyknosis、fragmentation in the 0.4 mCi group. In the 0.6 mCi group, a large of DRG cell became karyolysis and nuclear condensation. Mass DRG cell revealed karyolysis、nuclear condensation and degeneration. The ratio of positive cells(the nucleus was not in center or Cytoplasmic were 73.80±2.38% in 0.8 mCi group . Conclusion 1.DRG can be injured after implanting radioactive 125I around DRG, mainly characterized by necrosis, the intensity of which were increased as radiological dose increased.
