壳聚糖-左氧氟沙星长效缓释敷料的体外细胞毒性和抗菌效果研究

The Cytotoxic Dressing Effects and Antimicrobial Activity of Microspheres Containing Levofloxacin

摘要: 【摘要】目的  研究壳聚糖-左氧氟沙星长效缓释敷料的体外细胞毒性、抗菌效果以及肿瘤坏死因子(TNF)-α作为评价抗菌效果指标的可行性。方法  参照ISO 10993-5:2009评价该敷料的细胞毒性;制作染菌创伤白兔模型15只并分为3组,空白对照组用凡士林油纱包扎,阳性对照组用纳米银创伤贴包扎,实验样品组用壳聚糖-左氧氟沙星长效缓释敷料包扎,于6 h、1 d、4 d、7 d,采用活组织培养法检测3组的菌落数,用双抗体夹心法检测TNF-α含量,与菌落数实验结果进行比较。结果  该敷料体外细胞毒性反应为0级;实验样品组4个时点的菌落数均低于空白对照组,在6 h、1 d、4 d时,实验样品组均低于阳性对照组(P < 0.05);实验样品组的TNF-α含量在6 h、1 d、4 d、7 d时低于空白对照组,在6 h、1 d时显著低于阳性对照组(P < 0.05)。结论  壳聚糖-左氧氟沙星长效缓释敷料无细胞毒性,具有长效抗菌能力,TNF-α不能作为评价抗菌效果的指标。

Abstract: Objective   To study the antibacterial effect of wound dressing with microspheres containing levofloxacin in the live animal experiments and it’s cytotoxicity. Methods    According to ISO 10993-5:2009, study chitosan - cytotoxic effects of wound dressing with microspheres containing levofloxacin; vaseline gauze as a blank control, nano-silver trauma attached to experimental, the dressing as the experimental group, comparing different dressings after covering the wound, the number of colonies of the wound tissue, tumor necrosis factor (TNF-α) levels. Results    The vitro cytotoxic response of the dressing was 0/0. the number of colonies in 0.25,1,4,7 D were 96 ± 9 × 105CFU / g, 435 ± 235 × 105CFU / g, 5 ± 3 × 105CFU / g, 18 to 0; ± 9 × 105CFU / g; TNF-α levels in 0.25,1,4,7 D were 44.37 ± 3.39 Pg / mL, 41.45 ± 3.28 Pg / mL, 36.07 ± 3.63 Pg / mL, 32.91 ± 5.74 Pg / mL. Conclusion     the wound dressing was no significant cytotoxicity. with the growth of administration time, reduce the number of colonies and TNF-α, the wound dressing is a long-lasting antibacterial ability of medical dressings.
