鞣花酸抑制黑素细胞黑素合成及黑素传递的实验研究

Effects of ellagic acid on melanogenesis and melansome transfer

摘要: 目的:观察鞣花酸(Ellagic acid)对黑素细胞(melanocyte, MC)合成黑素以及传递黑素的影响并探讨其机制。方法:分别纯化培养来自人包皮的表皮黑素细胞和角质形成细胞(keratinocyte, KC),传第2代后以1:10的比例将两细胞接种到3cm×3cm的小培养皿中,单独或混合培养的细胞经高中低(100、10、1μg/ml)三种浓度的鞣花酸干预48 h后,分别检测干预前后黑素细胞中黑素含量、酪氨酸酶活性的变化以及流式法检测混合培养细胞中黑素传递的情况,以10nM烟酰胺为阳性对照。结果:鞣花酸呈浓度依赖性减少了黑素细胞黑素含量及酪氨酸酶活性。鞣花酸同时具有抑制黑素传递的作用。结论:鞣花酸可抑制黑素合成及黑素传递,或可作为一种皮肤美白剂应用。

Abstract: Objective To study if ellagic acid has preventing abilities on human epidermal melanocyte melanogenesis and melanin transfer in different concentration. Methods Our present investigated ellagic acid on, employing monolayer cultures of human melanocytes and co-cultures of human melanocytes and keratinocytes, melanogenesis and melanin transfer by Flow Cytometer technique. Results: Ellagic acid suppressed melanin synthesis and tyrosinase activity in a dose-dependent manner. Melanin transfer was also inhibited by ellagic acid in a dose-dependence manner. Conclusion: Ellagic acid may be useful for skin-whitening cosmetic and the depigmenting effect of ellagic acid might be due to its down-regulation of melanogenesis and melanin transfer.
