高良姜素抑制肺癌细胞 A549 增殖和侵袭的作用和机制

摘要: 目的 探讨高良姜素抑制人肺癌细胞 A549 增殖和侵袭的作用及机制。方法 对数生长期的 A549 细胞预铺于 96 孔板中,经过不同浓度高良姜素(0、5、10、20、40、60、100 μmol/L)处理 24 h,四氮唑蓝盐(MTT 的改良方法,MTS)试验检测高良姜素对细胞增殖的影响,计算半数抑制浓度(IC50)并采用无毒浓度(抑制率<5%)和低毒浓度(抑制率<15%)进行后续实验;Transwell 小室检测高良姜素对细胞侵袭的影响;Western blot 方法检测高良姜素对细胞中生存素(Survivin)、p27、CD44、细胞间黏附分子(ICAM)、基质金属蛋白酶(MMP)-2/9 的表达以及磷脂酰肌醇 3-激酶(PI3K)磷酸化(p-PI3K)和 AKT 磷酸化(p-AKT)的影响。结果 随着高良姜素浓度的增加,A549 细胞增殖率呈逐渐降低趋势,IC50为 44.7 μmol/L。采用 10、20 μmol/L 进行后续实验。0、10 及 20 μmol/L 高良姜素组的侵袭率呈逐渐降低趋势(P<0.05)。0、10 及 20 μmol/L 高良姜素组除 p27 表达水平依次增高外,其他各蛋白表达水平均呈依次降低趋势(P<0.05)。结论 高良姜素可抑制人肺癌细胞 A549 的增殖和侵袭,是潜在的肺癌治疗药物,其机制可能与调节相关基因蛋白水平及抑制 PI3K 和 AKT 磷酸化有关。

Abstract: Objective To explore the effect of galangin on the inhibition of proliferation and invasion in human lung cancer A549 cell lines and its mechanism thereof. Methods MTS assay was employed to detect the ability of cell proliferation with different concentrations (0, 5, 10, 20, 40, 60 and 100 μmol/L) of galangin after A549 cells were cultured in 96 well-plate. Transwell assay was employed to detect the ability of cell invasion. Western blot assay was employed to detect the protein expression levels of Survivin, p27, CD44, ICAM, MMP-2/9 and the phosphorylation of PI3K and AKT. Results The cell proliferation rate of A549 cell line was suppressed with increased concentration of galangin treatment, and IC50 was 44.7 μmol/L. The concentrations of 10 μmol/L and 20 μmol/L were used for the following study. The ability of cell invasion was decreased with 0, 10 and 20 μmol/L concentrations of galangin treatment (P<0.05). The protein expression of p27 was increased and the expressions of other proteins were decreased with 0, 10 and 20 μmol/L galangin treatmenmt (P<0.05).Conclusion Galangin can significantly inhibit the cell proliferation and invasion of human lung cancer A549 cell line, and which is a potential anti-lung cancer drug. The mechanisms may be related with the regulated related gene expression and the inhibited phosphorylation of PI3K and AKT.
