Panx1在非小细胞肺癌组织中的表达及临床意义

Expression and clinical significance of Panx 1 in non-small cell lung cancer

摘要: 目的 研究Panx1(Pannexin1)在人非小细胞肺癌(NSCLC)中的表达情况,并分析其临床意义。方法 应用实时荧光定量PCR法检测25例非小细胞肺癌组织及配对的癌旁组织中Panx1 mRNA的表达。应用免疫组织化学染色法检测30例非小细胞肺癌组织、配对的癌旁组织及10例正常肺组织中Panx1蛋白的表达,分析其表达与临床病理特征的关系。结果 NSCLC中Panx1 mRNA和Panx1蛋白的表达水平均高于癌旁组织,差异有统计学意义(P<0.05)。不同年龄、性别、胸膜侵袭情况、病理分级分组的NSCLC患者组织中Panx1蛋白阳性表达率比较差异无统计学意义(P>0.05),TNM临床分期Ⅲ~Ⅳ期组的Panx1阳性表达率高于Ⅰ~Ⅱ期组,有淋巴结转移组的Panx1阳性表达率高于无淋巴结转移组,差异均有统计学意义(P<0.05)。结论 Panx1在NSCLC组织中显著高表达,而在癌旁组织中低表达,提示Panx1可能在NSCLC的发生、发展及转移侵袭中起重要作用。

Abstract: To investigate the expression of Pannexin1 (Panx1) in human non-small cell lung cancer(NSCLC), and to analyze the clinical significance of Panx1 expression. Methods Real-time quantitative PCR was used to detect the expression of Panx1 mRNA in 25 NSCLC and matched paracancerous samples. The expressions of Panx1 protein were detected by immunohistochemical staining in 30 samples of NSCLC and matched paracancerous tissues, and 10 samples of normal lung tissues. The relationship between its expression and clinicopathological features was analyzed.Results The expression levels of Panx1 mRNA and Panx1 protein were significantly higher in NSCLC than those in adjacent tissues (P<0.05). There were no significant differences in positive expression rates of Panx1 between different age,gender, pleural invasion and pathological grading groups of NSCLC patients (P>0.05). The positive expression rate of Panx1 was higher in the TNM clinical stage Ⅲ-Ⅳ group than that in the Ⅰ-Ⅱ group. The positive expression rate of Panx1 was significantly higher in the lymph node metastasis group than that in the no lymph node metastasis group (P<0.05).Conclusion Panx1 is highly expressed in NSCLC tissues, but low in normal lung tissues or adjacent tissues, suggesting that Panx1 may play an important role in the occurrence, development and metastasis of NSCLC.
