子宫内膜样腺癌组织中 NF-κB p65与ERα的表达及意义

The expression and significance of NF-κB p65 in endometrioid carcinoma and its relationship with ERα

摘要: 本研究采用免疫组织化学SP方法检测不同类型子宫内膜组织中NF-κB p65蛋白和ERα蛋白的表达,旨在探讨NF-κB p65蛋白在不同类型子宫内膜组织腺上皮细胞中的表达及其与ERα蛋白之间的关系。结果发现NF-κB p65蛋白在正常子宫内膜组(10例)、子宫内膜增生症组(60例)以及子宫内膜样腺癌组(20例)中核表达率逐渐增高,且三组间差异具有显著性(p<0.05),内膜样腺癌组NF-κB p65蛋白的表达高于增生症组及正常内膜组,增生症组高于正常内膜组(p均<0.05);在增生症中,非典型增生高于单纯性增生及复杂性增生,复杂性增生高于单纯性增生(p均<0.05);ERα蛋白则在单纯性增生、复杂性增生、非典型增生子宫内膜和子宫内膜样腺癌组织中的表达逐渐降低,且具有统计学意义(p<0.05)。二者的表达在子宫内膜样腺癌组织中呈负相关性(r= -0.491,p=0.028)。最后,我们认为NF-κB p65蛋白在子宫内膜组织腺上皮细胞核中的表达率逐渐增加可能促进了子宫内膜样腺癌的形成和进展,ERα在子宫内膜样腺癌发展过程中表达逐渐下降,二者之间可能通过交互调节作用(cross-talk)而相互抑制。

Abstract: We used immunohistochemical SP methods to study the expression of NF-κB p65 and ERα in 10 cases of normal endometrium, 60 cases of endometrial hyperplasia, and 20 cases of endometrioid adenocarcinoma, aiming to investigate the expression of NF-κB p65 in different tissues of endometrial epithelial cells and its relationship with ERα. We observed that the expression of NF-κB p65 gradually raised from normal endometrium、endometrial hyperplasia to endometrioid adenocarcinoma , with significant difference among these three groups(p<0.05); and the expression of ERα gradually reduced from simple、complex、atypical hyperplasia to endometrioid adenocarcinoma, with significant difference among them(p<0.05). The overexpression of NF-κB p65 and the expression of ERα in endometrioid adenocarcinoma showed negative correlation (r= -0.491,p=0.028). In the end, we concluded that the increased expression of NF-κB p65 may play an important role in endometrioid carcinogenesis and its development, there maybe a molecular mechanism of cross-talk between NF-κB p65 and ERα.
