清醒大鼠高胰岛素-正血糖钳夹技术的建立和应用

Establishment and Application of Hyperinsulinemic-Euglycemic Clamp Technique in the Conscious Rats

摘要: 目的 建立大鼠高胰岛素-正血糖钳夹模型, 并探讨短期输注脂肪乳对清醒状态下大鼠葡萄糖输注率(GIR) 的影响。方法 SPF级雄性SD大鼠10只, 随机数字表法分为2组, 每组5只。采用颈动、 静脉插管技术, 分别经颈静脉给予SD大鼠输注20%脂肪乳 (脂肪乳组) 和5%葡萄糖6 h (对照组), 行高胰岛素-正血糖钳夹实验测定血浆游离脂肪酸(FFA) 和GIR。结果 与葡萄糖输注的对照组比较, 脂肪乳组大鼠输注20%脂肪乳6 h后, 血浆FFA水平升高到对照组的17.6倍 (P < 0.01), GIR下降了27% (P < 0.001), 胰岛素抵抗非常明显。结论 成功建立大鼠高胰岛素-正血糖钳夹技术, 给大鼠输注脂肪乳可导致明显的胰岛素抵抗。

Abstract: Objective: To establish hyperinsulinemic- euglycemic clamp technique in conscious rats, and to explore the effect of acute infusion of lipid on insulin sensitivity of rats. Methods: Rats were catheterized in the right jugular vein and left carotid artery, and underwent a hyperinsulinemic- euglycemic clamp(HEC) with infusion of lipid for 6 hours, in contrast with continuing infusion of 5% glucose. The glucose infusion rate (GIR) and plasma FFA were measured, respectively. Results: As expected, FFA concentration increased by 17.6-fold, and glucose infusion rates (GIR) was reduced by 27% in the Intralipid group compared to the controls with the 5% glucose group. Conclusion: A rat model of insulin resistance has been successfully esabished by intravenous intralipid infusion, which can be confirmed by the HEC technique.
