芦根乙醇提取物对糖尿病小鼠肝糖原含量及糖原合成酶的影响

Rhizoma Phragmitis ethanol extract in diabetic mice liver glycogen content and glycogen synthetase expression effect

摘要: 目的 探讨芦根乙醇提取物对链脲佐菌素 (STZ)诱导的糖尿病小鼠肝糖原含量及糖原合成酶 (GS)的影响。方法 正常对照组小鼠10只, 糖尿病小鼠随机分为模型对照组、 高剂量组和低剂量组,每组10只。应用组织化学糖原(PAS) 染色法检测各组小鼠肝糖原含量, 应用逆转录聚合酶链反应(RT-PCR) 和蛋白质免疫印迹法(West?ern-blot)法检测各组实验小鼠肝脏GS mRNA和蛋白的表达。结果 PAS染色后模型对照组肝糖原含量明显减少,低剂量组、 高剂量组较模型对照组明显增多 (P<0.01), 其中高剂量组最为明显; 模型对照组GS mRNA和GS蛋白均低于其他3组, 低剂量组低于高剂量组 (P<0.05)。结论 芦根乙醇提取物对糖尿病小鼠肝糖原含量具有正向促进作用, 其机制可能是通过提高肝糖原合成酶的表达而起作用。

Abstract: Abstract:objective To study Rhizoma Phragmitis ethanol extract influence on STZ induced diabetic mice liver glycogen content and glycogen synthetase. Methods Application periodic aci-Schiff reaction assay groups of laboratory mice liver glycogen content, and application reverse transcription polymerase chain reaction and Western blot assay Glycogen synthetase the expression of mice liver. Results After PAS staining the hepatic glycogen content decreased significantly after the model control group, low-dose group, high-dose group compared with model control group increased significantly (P<0.01), high-dose group was most pronounced among them; after RT-PCR, GS mRNA is lower than other three groups of model control group, low-dose group is lower than the high-dose group; after Westen blot after test model GS protein expression in the control group was lower than other three groups, low-dose group is lower than the high-dose group. Conclusions It is the dose-dependent regulation the Rhizoma Phragmitis ethanol extract influence on STZ induced diabetic mice liver glycogen, and raising the liver glycogen synthetase expression may be one of the role of the mechanism.
