降钙素和1,25维生素D拮抗牙周炎大鼠牙槽骨丧失的疗效观察

摘要: 目的 探讨降钙素与1,25维生素D对牙周炎导致大鼠牙槽骨丧失的影响。方法 雄性Wistar大鼠125只随机分为空白对照组 (A组)、 牙周炎对照组 (B组)、 降钙素治疗组 (C组, 降钙素溶于生理盐水, 2 μg/kg, 皮下注射)、1,25维生素D治疗组 (D组, 维生素D溶于花生油中, 2 μg/kg, 灌胃)、 降钙素和1,25维生素D联合治疗组 (E组), 每组25只。用正畸结扎丝结扎上颌磨牙牙颈部加高糖软食的方法建立牙周炎大鼠模型。在第2、 4、 6和8周末各组处死5只大鼠, 测量双侧上颌第一磨牙牙周探诊深度 (PD), 观察牙槽骨水平; 第8周末各组采取10只大鼠腹部降主动脉血,用酶联免疫吸附试验法 (ELISA) 检测血清中的骨碱性磷酸酶 (BALP) 及骨钙素 (OC) 的浓度。结果 4周、 6周、 8周时E组大鼠的PD低于B、 C、 D组 (P<0.05)。6周、 8周时E组大鼠牙槽骨丧失水平低于B、 C、 D组 (P<0.05)。第8周时E组BALP及OC浓度显著高于B、 C、 D组 (P<0.05)。结论 降钙素和1,25维生素D均能部分拮抗牙周炎导致的牙槽骨丧失, 二者联合应用效果更佳。

Abstract: Abstract Objective:The goal of this study was to investigate the effect of 1,25 Vitamin D and Calcitonin on experimental periodontitis through the animal experiment.Methods:40 male Wistar rats were allocated to four experimental groups according to the treatment received during 8 weeks: periodontitis group(group A), calcitonin (group B), 1,25 Vitamin D(group C), 1,25 Vitamin D plus calcitonin(group D).Eight weeks later,the animals were sacrificed and the specimens were prepared for histological analysis.clinical assessment included probing depth(PD) and aleolar bone loss(ABL).Histological examination included periodontal membrane fiber and alveolar bone inflammatory.The serum BALP and OC were measured by ELISA method.Results:Intergroup comparisons demonstrated that group D might reduce the probing depth and the alveolar bone loss was decreased compared with group A,B and C (P<0.05).The serum BALP and OC in group D was higher than group A , B and C.Conclusions:The present study demonstrates that after taken 1,25vitaminD and Calcitonin for 8 weeks experimental periodontitis rats could decreased alveolar bone loss.
