大黄素对人白蛋白诱导人肾小管上皮细胞转分化的影响

摘要:

【摘要】 目的探讨大黄素对人白蛋白诱导的人肾小管上皮细胞(HK-2)发生转分化的影响。方法体外培养HK-2细胞,按处理方式分为空白对照组、大黄素对照组、白蛋白诱导组、大黄素干预组4组。分别于0h、12h、24h、48h用倒置显微镜下观察其细胞形态变化,同时RT-PCR法检测各样本α-SMA、E-cadherin、FNmRNA的表达水平。结果空白对照组、大黄素对照组HK-2细胞形态为圆形,融合后呈鹅卵石样。白蛋白诱导组12、24及48h光镜下可见细胞胞体拉长变细,细胞之间间隙增大。大黄干预组12、24、48h后可见细胞形态改变与白蛋白诱导组相似,但改变程度较小。空白对照组和大黄素对照组表达α-SMA、E-cadherin、FN的mRNA水平差异无统计学意义(均P>0.05)。与空白对照组比较,24和48h白蛋白诱导组、大黄素干预组α-SMA、FN的mRNA随时间延长表达逐渐增强,E-cadherinmRNA逐渐减弱。24和48h大黄素干预组上述指标的表达改变弱于白蛋白诱导组(均P<0.05)。结论大黄素可以抑制人白蛋白诱导的HK-2细胞转分化,延缓肾间质纤维化的进展。

Abstract: Objective To explore the effect of emodin on the transdifferentiation of human renal tubular epithelial cell to mesenchymal induced by human albumin. Methods Human kidney proximal tubular epithelial cell line (HK-2) was stimulated by human albumin(5mg/ml)in vitro. Morphological changes of the HK-2 cells were observed by an inverted phase contrast microscope. The expressions of α-SMA, E-cadherin, FNmRNA were detected by reverse transcription polymerase chain reaction(RT-PCR). Results Albumin up regulated α-SMA, FN mRNA expression in HK-2 cells in a time-dependent manner and E-cadherin mRNA was down-regulated. Emodin repressed the effect of albumin on them when using a certain concertration emodin. Conclusion Emodin inhibit the transdifferentiation of human renal tubular epithelial cell to mesenchymal induced by human albumin., slow down the progression of renal interstitial fibrosis.
