胰腺癌细胞中GDNF对MMP-9表达的影响

Influence Of GDNF To The Expression Of Matrix Metalloproteinase 9 In Pancreatic Carcinoma

摘要: 目的探讨胰腺癌细胞中胶质细胞源神经营养因子(GDNF)对基质金属蛋白酶(MMP)-9表达的影响。方法用DMEM培养基进行人胰腺癌细胞株MIAPaCa-2和Capan-2细胞培养,利用RT-PCR、Westernblot和ELISA法研究胰腺癌细胞中GDNF对MMP-9mRNA及蛋白表达的影响。结果 RT-PCR实验发现胰腺癌细胞中GDNF能上调MMP-9mRNA的表达,Westernblot法、ELISA法检测结果表明胰腺癌细胞中GDNF能上调MMP-9蛋白的表达,且于GDNF浓度为100μg/L,作用48h时达最大值。结论 GDNF具有上调胰腺癌细胞MMP-9表达的作用,从而增强胰腺癌细胞的侵袭能力。

Abstract: Abstract: Objective To investigate the influence of Glial Cell Line–Derived Neurotrophic Factor(GDNF) to the expression of Matrix Metalloproteinase 9(MMP-9) in pancreatic carcinoma. Methods Make use of RT-PCR 、Western blot and Elisa to investigate the effect of GDNF to the expression of MMP-9 mRNA and protein in MIA PaCa-2 and Capan-2 human pancreatic carcinoma cell line. Results We found that GDNF can up-regulate the expressions of MMP-9 mRNA and its protein in a dose-dependent manner in RT-PCR and Elisa experiments. Conclusions GDNF has the effect to stimulate the expression of MMP-9 and thereby strengthen the anility of invasion in panereatic carcinoma. Key Words: Pancreatic Carcinoma; GDNF; MMP-9; Expression
