多西环素对乳腺癌小鼠移植瘤生长及转移的影响

Effects of Doxycycline on Tumor Growth and Metastasis in Breast Cancer Xenografts in Mice

摘要:

【摘要】目的 通过检测多西环素对小鼠乳腺癌肺转移模型(BCML-TAⅡ99)中基质金属蛋白酶-9(MMP-9)及金属蛋白酶组织抑制因子1(TIMP-1)蛋白表达水平,揭示其对该肿瘤生长及转移的影响。方法建立BCML-TAⅡ99 模型,应用免疫组织化学SP法检测MMP-9和TIMP-1蛋白在多西环素给药组与对照组肿瘤组织中的表达情况,并进行比较。结果与对照组相比,给药组肿瘤转移率明显低于对照组;MMP-9蛋白在给药组表达下调,而TIMP-1蛋白表达上调,差异具有统计学意义(P<0.05),且MMP-9与TIMP-1表达呈负相关(P<0.05)。结论多西环素可通过调节MMP-9及TIMP-1蛋白表达水平,对BCML-TAⅡ99模型中肿瘤生长及转移发挥抑制作用。

Abstract:

[Abstract] Objective To investigate the effects of doxycycline on the expressions of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metaloproteinase-1(TIMP-1) in breast cancer lung metastasis model mice (BCML-TAⅡ 99), and the mechanism thereof. Methods The BCML-TAⅡ99mouse model was established. The immunohistochemical staining was performed to detect the expressions of MMP-9and TIMP-1in BCML- TAⅡ99model mice and controls. Results The metastasis rate was significantly lower in treatment group than that of control group. The expression of MMP-9 was significantly reduced, but the expression of TIMP-1was significantly increased, in treatment group compared with those of control group (P<0.05). Conclusion Doxycycline can inhibit tumor growth and metastasis in breast cancer with1ung metastasis model mice by regulating MMP-9and TIMP-1expressions.
