YD383和YD439对IL-4诱导的BJAB细胞CD23表达的影响

The Effect of YD383 and YD439 on the Expression of CD23 Induced by IL-4 in BJAB Cells

摘要: 摘要 目的:观察两种小分子化合物YD383,YD439 对IL-4诱导的BJAB 细胞 CD23分子表达的影响,初步探讨两种化合物对JAK/STAT6信号传导通路的抑制机理。方法:将不同剂量的化合物YD383和YD439加入对数生长期的BJAB细胞作用半小时,终浓度为20ng/mL 的IL-4共同孵育3小时后,收集细胞并加入PE标记的鼠抗人CD23抗体,流式细胞仪检测各细胞群体CD23分子的表达情况。结果:化合物YD383和YD439均能降低由IL-4诱导的BJAB细胞CD23分子的表达,且随着化合物剂量的增大,CD23分子的表达水平逐渐降低,。两种化合物各自不同的剂量组间,CD23分子表达 的差异均有统计学意义(P<0.05)。结论:化合物YD383和YD439对IL-4诱导的CD23分子的表达均有明显的抑制作用,并且具有剂量依赖性。

Abstract: Abstract Objective: To examine the effect of compounds YD383 and YD439 on the expression of CD23 induced by IL-4 in BJAB cells. Methods: The BJAB cells of exponential growing were treated with YD383 or YD439 in different dose for 30 minutes. Then cells were incubated with IL-4 (20ng/ml) for 3 h. Cells were collected and mixed with PE-lableled CD23 antibody. The expression of CD23 in different conditions was detected by Flow Cytometry. Results: The expression of CD23 induced by IL-4 in BJAB cells were significantly inhibited by YD383 and YD439 in a dose dependent manner (P<0.05) Conclusion: YD383 and YD439 can inhibit the expression of CD23 induced by IL-4 in BJAB cells. The expression of CD23 decreased in time-dependent manner.
