大鼠心肌梗死后非梗死区基质重构及相关调节因子变化的研究

Experimental Study on Matrix Reconstruction in Non-infarcted area and Related Regulating Factors Changing with Time after Myocardial Infarction in Rats

摘要: 摘要 目的:观察大鼠急性心肌梗死后非梗死区心肌基质重构相关调节因子随时间变化的关系,探讨大鼠心肌梗死后基质重构的机制;方法:结扎左冠状动脉前降支制作大鼠心肌梗死模型,分别于术后1、3、7、14和28天处死动物,采用Masson染色检测非梗死区心肌胶原容积分数(collagen volume fraction,CVF);明胶酶谱法检测MMP-2、TIMP-2活性;实时荧光定量PCR法检测NF-ΚB、TNF-αmRNA表达。结果:与假手术组相比较,心肌梗死组大鼠非梗死区CVF随梗死后时间的延长而增加,28天最明显(P<0.01); MMP-2 、TIMP-2活性随时间延长而增加,两者在梗死后14天达高峰,后有所下降,28天时活性仍高于假手术组(P<0.05);TNF-α的表达于梗死后7天达高峰(P<0.01),后有所下降,持续到28天,仍高于假手术组(P<0.05);NF-κB的表达活性随梗死时间的延长而增加(P<0.05,P<0.01)。结论:大鼠急性心肌梗死后,心肌非梗死区MMP-2/TIMP-2及相关调节因子活性改变具有明显时间依从性,是导致非梗死区基质重构的机制之一。

Abstract: Objective: To observe matrix reconstruction related regulating factors changing with time and research matrix reconstruction mechanism after acute myocardial infarction in rats. Methods: Ligating left anterior descending coronary artery of rats to make myocardial infarction model, the rats were killed on 1 day, 3 days, 7 days, 14 days, 28 days separately after operation, using Masson dyeing to detect collagen volume fraction (CVF) in non-infarcted area; Gelatinases spectrum method to detect MMP-2, TIMP-2 activity; Real-time fluorescence quantitative polymerase chain reaction (PCR) to detect mRNA expression of NF-ΚB and TNF-α.Results: Compared with Sham operation group, CVF in non-infarcted area increased with time in infarction group, most obviously 28 days after infarction (P<0.01); activity of MMP-2 and TIMP-2 increased with time, both of them reached peak 14 days after infarction then decreased, but remained more active than Sham operation group by 28 days (P<0.05); expression of TNF-αreached peak 7 days after infarction then decreased, continuing to 28 days after infarction, still higher than Sham operation group (P<0.05); expression of NF-κ B increased with infarction time(P<0.05, P<0.01). Conclusions: Activity changes of MMP-2、TIMP-2 and related regulating factors complied obviously with infarction time, it might be an important mechanism of matrix reconstruction in non-infarcted area after acute myocardial infarction in rats.
