人脐带间充质干细胞与大鼠激活态雪旺细胞联合培养的实验研究

Study on co-culture of human Umbilical Cord Mesenchymal Stem Cells and rats’ Activated Schwann Cells

摘要: 目的: 探讨体外分层共培养条件下大鼠激活态雪旺细胞对人脐带间充质干细胞生长及分化的影响。方法: 分离、培养人脐带间充质干细胞和大鼠激活态雪旺细胞,通过milicell小室实现两者体外共培养,nestin、NF-200免疫荧光染色分析干细胞分化情况。结果: milicell小室分层共培养条件下,hUCMSCs逐渐出现类似神经细胞的形态,对照组形态无明显变化。共培养后MSCs表达神经干细胞标志物Nestin及神经元标志物NF200,对照组MSCs低表达Nestin而不表达NF200。结论: 人脐带间充质干细胞有向神经细胞分化的能力,分层培养条件下大鼠激活态雪旺细胞可促进其神经方向分化。

Abstract: Abstract Objective:To study the influence of growth and differentiaion of human Umbilical Cord Mesenchymal Stem Cells when co-cultule with Activated Schwann Cells in vitro. Methods:Isolated and cultured human Umbilical Cord Mesenchymal Stem Cells and rats’ activated Schwann cells, co-culture these two types of cells by milicell inserts, Immunofluorescence staining of nestin and NF-200 were using to analysis the differentiation of hUCMSCs. Results:In milicell inserts layered co-culture conditions, hUCMSCs showed neuronal morphology gradually which was not happened in the control group; hUCMSCs expressed neural stem cell marker Nestin and neural marker NF200,while in the control group expression of Nestin was in a low level and NF200 was negative. Conclusion:hUCMSCs was able to transdifferentiate into neural cell, and neural differentiation can be induced by coculture with Activated Schwann Cells .
