Stathmin沉默抑制鼻咽癌5-8F细胞的恶性表型

Stathmin silence suppressing malignant phenotype ofnasopharyngeal carcinoma 5-8F cell line

摘要: 【摘要】 目的  探讨stathmin沉默对鼻咽癌5-8F细胞株的生物学作用。方法  实验设空白对照组(未转染载体的5-8F细胞),阴性对照组(转入pGenesil-1.1-HK质粒表达载体),stathmin沉默组(转入pGenesil-1.1-STM质粒表达载体),siRNA重组表达载体采用脂质体转入鼻咽癌5-8F细胞,然后进行G418筛选,挑取单克隆扩大培养,获得稳定转染的5-8F细胞株。采用MTT实验、流式细胞仪分析细胞的增殖和周期;裸鼠移植瘤实验分析细胞的成瘤性。结果  Stathmin沉默组5-8F细胞增殖受抑制,其抑制率(33.67±2.52)%明显高于阴性对照的(8.13±0.60)%和空白对照组的0,差异有统计学意义(P<0.05);Stathmin沉默组细胞阻滞于G2期,G2期细胞为(15.77±0.55)%,高于阴性对照组的(11.32±0.79)%和空白对照组的(10.90±0.92)%(P<0.05);Stathmin沉默组裸鼠移植瘤质量为(0.32±0.21)g,低于阴性对照组的(1.10±0.40)g和空白对照组的(1.52±0.30)g(P<0.05)。结论  Stathmin沉默能抑制鼻咽癌5-8F细胞的恶性生物学表型。

Abstract: Abstract   Objective   Exploring biological effects of stathmin silencing on nasopharyngeal carcinoma 5-8F cell line. Methods    Cell proliferation and cell cycle were analyzed by MTT experiment and Flow Cytometry. Cell tumorigenicity was analyzed by nude mouse xenograft experiment. Results    The cell proliferation inhibition rate of stathmin-silenced 5-8F cells was significantly higher than that of negative and blank control group cells (p<0.01). The cell cycle of stathmin-silenced 5-8F cells was significantly arrested in phase G2 compared with that of negative and blank control group(P<0.05). The transplanted tumor weight of stathmin-silenced 5-8F cells in nude mice was (0.32±0.21)g,significantly lower than that of negative control group (1.10±0.40)g and blank control group(1.52±0.30)g(P<0.05). Conclusion   Stathmin-silencing inhibits malignant phenotype of nasopharyngeal carcinoma 5-8F cell line.
