线粒体分裂在甲状腺鳞癌SW579 细胞增殖、 凋亡以及侵袭中的作用

Effects of mitochondrial fission in the cell proliferation ,apoptosis and invasion of thyroid squamous cell carcinoma cell line SW579

摘要: 摘要: 目的 研究线粒体动力学蛋白 Mfn2 和 Drp1 在甲状腺鳞癌 SW579 细胞中的表达和线粒体分裂抑制剂-1 (Mdivi-1) 对 SW579 细胞增殖、 凋亡及侵袭能力的影响。方法 采用 RT-PCR 检测 Mfn2 和 Drp1 mRNA, Western blot 检测 Mfn2 和 Drp1 蛋白在 Nthy-ori 3-1 和 SW579 两种细胞中的表达; 然后将 SW579 分为对照组 (DMSO, 0.1%)和 Mdivi-1 低、 中、 高剂量组 (浓度分别为 15、 30、 45 μmol/L), 培养 16 h, 采用 MTT 法检测细胞增殖能力的变化, 用荧光分光光度计检测线粒体膜电位的变化, 用 RT-PCR 检测细胞色素 C (Cyt C) 和 Caspase-3 mRNA 的变化, Western blot 检测 Cyt C 和 Caspase-3 蛋白的变化, 用 Transwell 小室侵袭实验检测侵袭能力的变化。结果 与 Nthy-ori 3-1 细胞系相比, SW579 细胞系中 Mfn2 mRNA 和蛋白表达明显降低, Drp1 mRNA 和蛋白表达明显升高 (P<0.01); 与对照组相比, Mdivi-1 各组 SW579 细胞存活率和线粒体膜电位均明显降低, Cyt C 和 Caspase-3 的 mRNA 及蛋白表达均明显增加, 侵袭能力明显减弱 (均 P<0.01), 并且呈药物浓度依赖性。结论 甲状腺鳞癌 SW579 细胞中存在线粒体动力学异常, Mdivi-1可以抑制此细胞的增殖和侵袭, 并能诱导其凋亡。

Abstract: ObjectiveTo detect the changes of mitochondrialdynamicsproteins(Mfn2 and Drp1) in cell line SW579, and the effects of Mdivi-1 on cell proliferation,apoptosis and invasion of cell line SW579. Methods In SW579 and Nthy-ori 3-1, the expression levels of Mfn2 and Drp1 were determined by western blot, the Mfn2 and Drp1 mRNA expression levels were measured by RT- PCR .Then, SW579 cells were divided into control group(DMSO, 0.1%)and Mdivi-1 group (different concentrations of Mdivi-115,30 and 45 μmol/L). SW579 cells were treated with Mdivi-1 for 16 h。Then the ability of cell proliferation was detected using MTT assay,the mitochondrial membrane potential was detected by fluorescence spectrophotometer, the expression levels of cytochrome C and Caspase-3 were measured by Western blot, the Cyt C and Caspase-3 mRNA expression levels were determined by RT- PCR. the ability of invasion in each group was measured with Transwell assays. Results Compared with Nthy-ori 3-1,The mRNA and protein expression of the Mfn2 was remarkably decreased, whilethe mRNA and protein expression of the Drp1 was significantly increased in SW579 (P<0.01). Compared with the control group, the cell survival rates and the mitochondrial membrane potential of SW579 weredecreased dramatically(P<0.01),the mRNA and protein expression of the cytochrome C and Caspase-3 were increased dramatically (P<0.01), the capability of invasion was markedly decreased in the Mdivi-1 group (P<0.01) , which are all in a concentration-dependent manners (P<0.01). Conclusion Abnormal mitochondrial dynamics may be involved in thyroid squamous cell carcinomaSW579 cells; Mdivi-1 can inhibit the cell proliferation as well as invasion, and also induce apoptosis.
