低氧刺激GES-1细胞后 MICA基因在mRNA和蛋白水平的表达

Expression of MICA at the mRNA and protein levels in Human Gastric Epithelial cell Strain-1 of Hypoxia

摘要: 【摘 要】目的:研究低氧对体外培养胃上皮细胞株GES-1中MICA 基因表达的影响。方法:建立GES-1细胞体外低氧培养模型。在缺氧处理后(12h、24h、72h) ,RT -PCR 和免疫细胞化学技术分别检测MICA 基因和蛋白的表达变化。结果:随缺氧增加,免疫细胞化学及RT-PCR证实MICA 蛋白的表达与MICAmRNA 的表达同步, 且随着缺氧时间的增加呈时间效应关系。且表达量随缺氧时间而升高,Pearson级差相关分析显示MICA mRNA和蛋白水平表达显著正相关。结论:低氧氧可以显著增加胃上皮细胞株GES-1中MICA 基因的表达。

Abstract: 【Abstract】Objective :To investigate the effect of hypoxia on MICA expression in GES-1 its mechanism. Methods :Hypoxia culture model in vitro was established to test the expression of MICA in GES-1 cell line. After cultured in hypoxia circumstance for a given period of time (12hours ,24hours ,72hours ) ,MICA expression was tested in mRNA level and protein level by RT - PCR and immunocytochemistry analysis . Results: Cell survival was affected a little within 20 hours under hypoxia. MICA expressions increased after treatment of hypoxia. MICA protein expressions increased equally with mRNA confirmed by immunocytochemistry and RT - PCR and its positive correlation was found by Pearson product - moment correlation analysis. Conclusion :MICA gene and protein can be induced remarkably byhypoxia in GES-1.
