HIF-1α和STAT3分子表达与大肠癌淋巴转移的关系

摘要: 摘要 目的:探讨缺氧诱导因子(HIF-1α)、信号转导和转录活化因子3(STAT3)在大肠癌淋巴转移中的作用机制。方法:采用荧光实时定量PCR和免疫组织化学染色方法检测60例大肠癌组织及远癌切端大肠癌组织HIF-1α、STAT3和VEGF-C的mRNA转录水平和蛋白表达情况,同时以抗podoplanin单克隆抗体标记大肠癌微淋巴管计数淋巴管密度(LVD),分析上述指标与大肠癌淋巴结转移之间的相互关系。结果:大肠癌组织中HIF-1α、STAT3和VEGF-C分子的mRNA转录水平及蛋白表达水平明显高于远癌切端大肠癌组织(P<0.05),大肠癌组织中HIF-1α、STAT3与VEGF-C分子mRNA转录水平及蛋白表达水平呈正相关,有淋巴结转移患者LVD高于无淋巴结转移患者(P<0.05)。结论:大肠癌组织较远癌切端大肠组织有更为密集的淋巴系统,大肠癌组织中HIF-1α、STAT3、VEGF-C是调节大肠癌淋巴管形成的关键因子。

Abstract: ABSTRACT Objective: Study the mechanism of action of hypoxia inducible factor-1α(HIF-1α), signal transducers and activators of transcription 3(STAT3) in lymphatic metastasis of colorectal carcinoma. Methods: The levels of HIF-1α, STAT3 and VEGF-C mRNA transcription and protein expression of adenocarcinoma and surgical margin tissues gotten from 60 cases with CRC were determined by real-time quantitative PCR using fluorescence and immunohistochemistry, as well as lymph vessel density was counted using anti-podoplanin monoclonal antibody. And correlations of above indexes and lymphatic metastasis of colorectal carcinoma were analyzed. Results: The levels of HIF-1α, STAT3 and VEGF-C of tumor tissues were significant higher than that of surgical margin in either mRNA transcription or protein expression(P<0.05). The levels of HIF-1α, STAT3 and VEGF-C of tumor tissues were more significant relative not only in either mRNA transcription or protein expression but also in lymphatic metastasis group(P<0.05). Conclusion: The expression of lymphatic vessels system in colorectal carcinoma is higher than that in surgical marginal tissue. HIF-1α,STAT3 and VEGF-C are the key regulators in lymphatic vessel formation of colorectal carcinoma,and are closely related to lymphatic metastasis.
