内皮型一氧化氮合酶基因多态性与原发性高血压关系的研究

摘要: 目的: 探讨天津市汉族人群内皮型一氧化氮合酶(eNOS)基因G894T和27bp数目可变的串联重复序列(VNTR)多态性与原发性高血压的关系。方法:采用PCR-RFLP对350例原发性高血压患者和150例对照组eNOS第7外显子894位及第4内含子VNTR进行基因多态性分型;同时按照流行病学方法制作规范的调查表,调查内容包括吸烟,饮酒,体重指数等方面情况。结果:(1)eNOS 894位点EH组GT+TT基因型及T等位基因频率与对照组相比无显著型差异(P>0.05).(2)eNOS 第4内含子VNTR EH组aa+ab基因型及a等位基因频率显著高于对照组(P<0.05).(3) eNOs基因G894T和第4内含子VNTR对高血压的发生无协同作用 结论:eNOS基因G894T多态性与原发性高血压无关;而第4内含子VNTR多态性与原发性高血压存在相关性,两者无协同作用.

Abstract: Objective To investigate the association between G894T and 27bp varible number of tandem repeats(VNTR) polymorphism of endothelial nitric oxide synthase(eNOs) gene and essential hypertension(EH) Methods The PCR-RFLP method was used to detect genetypes of eNOS gene exon 7th 894 site and intron 4 th VNTR in350 patients with EH and 150 controls. Questionnaire refered to smoking,drinking, body mass index,etc was administered.Results There was no significant difference in frequencies of T allele and GT+TT genotypes in hypertension group than in control. The frequencies of a allele and aa+ab genotypes were significantly higher in hypertension group than in control. There was no significant synergistic effects of eNOS gene exon 7th 894 site and intron 4 th VNTR in EH.Conclusion The polymorphism of eNOS G894T was not associated with hypetension,while the polymorphism of intron 4 th VNTR was associated with hypertension,there was no significant synergistic effects.
