原白头翁素衍生物通过外源性途径诱导人乳腺癌MCF-7细胞凋亡

Mechanism of isomycin derivant on apoptosis of human breast carcinoma MCF-7 cell line

摘要: 摘要:目的:探讨原白头翁素衍生物诱导人乳腺癌MCF-7细胞株凋亡的作用机制,为开发新的抗肿瘤中药提供实验和理论依据。方法:采用体外培养方法培养MCF-7细胞;应用免疫细胞化学技术(SABC法)检测药物处理前后凋亡通路相关蛋白Fas,凋亡蛋白激酶Caspase-3、Caspase-8,细胞核转录因子NF-κB的表达变化并探讨其意义。结果:原白头翁素衍生物能上调Fas、Caspase-8、Caspase-3的表达,并能下调NF-κB的表达,与对照组相比较均有显著性差异。结论:原白头翁素衍生物通过外源性途径即死亡受体通路完成凋亡的启动和执行,另外抑制NF-κB信号转导途径也可能是促进MCF-7细胞凋亡的原因。

Abstract: Objective: To investigate the mechanism of isomycin derivant on anti-proliferation and inducing apoptosis of human breast carcinoma MCF-7 cell line in vitro. Methods: MCF-7 cells were cultured in vitro; We investigated the expression of Fas、Caspase-3、Caspase-8、NF-kB by immunohistochemitry and studied the possible mechanism of the apoptosis. Results: Immunocytochemistry showed that Isomycin derivant up-regulated the expression of Fas、Caspase-8、Caspase-3,and they were significantly different from control groups; Isomycin derivant also inhibited signal transduction pathway of NF-kB. Conclusions: Isomycin derivant induced apopsis via extrinsic or death receptor-mediated pathway; Inhibition of the signal transduction pathway of NF-kB may be the mechanism of inducing apoptosis.
