神经节苷脂对脑缺血再灌注大鼠海马 Fas 表达的影响

Effects of Monosialoganglioside on the Expression of Fas in Hippocampus of Cerebral Ischemia-Reperfusion Rats

【摘要】 目的  探讨单唾液酸神经节苷脂(GM)1对脑缺血再灌注大鼠海马Fas表达的影响。方法 72只SD大鼠随机分为假手术组(8只)、GM1治疗组(32只)、缺血再灌注组(32只)。GM1治疗组和缺血再灌注组依据缺血后再灌注的不同时间点再分为6h、12h、24h、3d共4个小组。应用免疫组织化学方法和Western blot方法检测各组大鼠海马Fas蛋白的表达。结果  免疫组织化学方法结果显示假手术组大鼠海马有一定量的Fas蛋白表达(0.17±0.02),而缺血再灌注组大鼠海马的Fas蛋白阳性表达显著升高,各时点分别为0.42±0.11、0.51±0.13、0.55±0.13及0.62±0.15,3d时表达最高;与相同时间点的缺血再灌注组比较,GM1治疗组的Fas蛋白阳性表达则明显降低,各时点分别为0.29±0.09、0.34±0.11、0.37±0.12、0.43±0.12(均P<0.05)。Western blot结果与此结果类似。结论 GM1很可能通过减少Fas蛋白的表达参与脑缺血再灌注损伤的神经保护作用。

Abstract: 0bjective   To study the expression of Fas in hippocampus of cerebral ischemia-reperfusion rat after monosialoganglioside was injected. Methods   72 SD rats were randomly divided into the sham group, ischemia-reperfusion group(I/R), monosialoganglioside group(GM1) at different time points (6h, 12h, 24h, 3d) after reperfusion on average. The expression of Fas in hippocampus of all groups was examined by immunochemistry and western blot methods. Result   There is a little of Fas positive expression in hippocampus of the rats in sham group, and the positive expression of Fas was increased significantly after reperfusion (P< 0.05), to the peak at 3d, compared with the rats of the I/R group at the same time point, the positive expression of Fas decreased significantly in GM1 group (P< 0.05) ; and the same result was obtain by western blot method. Conclusion   one of the mechanisms for monosialoganglioside participate the pathogenesis of cerebral ischemia-reperfusion injury might be downregulate the expression of Fas.
