脑胶质瘤组织中的细胞周期分析与意义

Analysis and significance of cell cycle phase in human glioma

摘要: 目的:探讨脑胶质瘤组织中细胞周期蛋白(Cyclin D1、Cyclin A、Cyclin B1)和核分裂特异性抗体(PHH3)的表达规律,方法:采用免疫组化S-P法检测Cyclin D1、Cyclin A、Cyclin B1、PHH3在59例脑胶质瘤组织(Ⅰ级10例、Ⅱ级15例、Ⅲ级15例、Ⅳ级19例)和37例正常脑组织中的表达情况。结果:Cyclin D1、Cyclin A、Cyclin B1、PHH3在正常脑组织中不表达。胶质瘤组织中Cyclin D1、Cyclin A、Cyclin B1、PHH3的表达阳性率分别为69.5%、69.5%、62.7%、84.7%。随着胶质瘤级别升高,Cyclin D1、Cyclin A、Cyclin B1、PHH3的平均标记指数升高,4级脑胶质瘤中Cyclin D1、Cyclin A、Cyclin B1、PHH3的阳性标记指数比较差异有统计学意义(P< 0.01)。结论:可通过免疫组织化学法分析细胞周期的数据,这有利于肿瘤分级,并与预后相关。

Abstract: Objective: To explore the expression and significance of Cyclin D1, Cyclin A, Cyclin B1 and PHH3 in human glioma, and study the correlation between parameters and clinicopathological features. Methods: Expression of Cyclin D1, Cyclin A, Cyclin B1 and PHH3 were analyzed by S-P immunohistochemical analysis in 59 cases of gliomas(gradeⅠ ten cases, grade Ⅱ fifteen cases, grade Ⅲ fifteen cases, grade Ⅳ nineteen cases) and 37 normal brain tissues. Results: Expression of Cyclin D1, Cyclin A, Cyclin B1 and PHH3 were not found in normal brain tissues. The positive expression rate of Cyclin D1, Cyclin A, Cyclin B1 and PHH3 in gliomas were 69.5%、69.5%、62.7% and 84.7% respectively. With the increasing of pathological grades, the mean labeling indexes for Cyclin D1、Cyclin A、Cyclin B1 and PHH3 in human gliomas increased accordingly. The difference of Cyclin D1、Cyclin A、Cyclin B1、PHH3 labeling index between four groups were significant(P<0.01). Conclusion: Cell cycle phase can be analyzed by immunohistochemical stain, and it is beneficial for tumor grading and helpful to predict the prognosis.
