内毒素血症大鼠肾脏水通道蛋白2等基因表达及肾脏结构功能的改变

Effect on the Expression of Related Protein of AQP2 and Alteration of Morphology and Function in Kidney of Endotoxemic Rats

摘要: 目的 探讨大鼠内毒素血症肾脏AQP2、ICAM-1、TNF-α mRNA和蛋白表达及肾脏结构功能的改变。方法 Wistar大鼠24只随机分为3组,每组8只:对照1(C1)组、对照2(C2)组、内毒素(L)组,模型制备成功12小时后,处死取材。观察肾组织病理学改变,检测肾脏功能学指标,RT-PCR、Western Blot及免疫组化方法测定AQP2、ICAM-1和TNF-α的表达。 结果: L组肾组织出现病理学损伤,功能学指标较C1、C2组受损明显,AQP2 mRNA及蛋白表达减少,ICAM-1、TNF-α mRNA及蛋白表达增多。 结论 大鼠内毒素血症可导致肾组织形态和功能损伤,AQP2表达减少,ICAM-1和TNF-α表达增多。

Abstract: Objective: To investigate the effects on the expression of related protein of AQP2 and alteration of morphology and function in kidney of endotoxemic rats. Methods: 24 rats were randomly divided into 3 groups (n=8 each): C1 group (sham control group), C2 group (saline and fat milk group), and L group (endotoxin and fat milk group). The morphology and function of kidney and the levels of AQP2, ICAM-1, and TNF-α with RT-PCR and Western blot were detected. Results: Endotoxemia led to morphological damage and functional impairment, and downregulation of AQP2 and upregulation of ICAM-1 and TNF-α in rat kidney. Conclusions: Endotoxemia led to renal damage, and decreased AQP2 level, and increased ICAM-1 and TNF-α level.
