Hhcy及B族维生素干预对大鼠脑血管ICAM-1表达的影响

摘要: 目的 探讨高同型半胱氨酸血症(hyperhomocysteinemia,Hhcy)及叶酸、维生素B12和维生素B6干预对大鼠脑皮层血管细胞间粘附分子-1(intercellular adhesion molecule-1,ICAM-1)表达的影响。方法 采用不同饮食结构及干预手段将40只雄性SD大鼠随机分为对照组、Hhcy组、治疗组和预防组,采用高效液相色谱(high performance liquid chromatogram,HPLC)荧光检测技术测定不同时期各组大鼠血浆总同型半胱氨酸(total Homocysteine,tHcy)水平,并于第12周末通过免疫组化法观察ICAM-1在大鼠脑皮层血管的表达情况。结果 第4周末大鼠Hhcy模型制备成功,而第12周末对照组、治疗组和预防组血浆tHcy水平无统计学差异;各组ICAM-1阳性血管个数差异均有统计学意义(P<0.05),其表达高低顺序依次是Hhcy组(3.23±0.58)>治疗组(1.54±0.36)>预防组(0.98±0.23)>对照组(0.35±0.31)。结论 Hhcy可以促进大鼠皮层脑血管ICAM-1表达,B族维生素可以有效降低血浆tHcy水平,并且对ICAM-1表达有一定的干预作用。

Abstract: Objective To observe the expression of ICAM-1 in the brain of rats with Hhcy or after the intervention of folic acid, Vitamins B12 and Vitamins B6. Methods 40 healthy male Sprague-Dawley rats were divided into 4 groups as:normal group, Hhcy group,treat group, prevent group by the way of different foods and different interventions.The plasma total homocysteine (tHcy) levels of rats were assayed by high-performance liquid chromatography method(HPLC) in different time; At the end of 12th week ,the expression of intercellular adhesion molecule-1 (ICAM-1) were detected by immunohistochemical method in the cerebral vessels of rats. Results At the end of 4th week, the Hhcy model were induced successfully; At the end of 12th week ,there were no statistical difference between the treat group,prevent group and normal group; there were significantly statistical differences between the number of masculine cerebral vessels in the 4 groups(P<0.05), the sequence is Hhcy group(3.23±0.58)>treat group(1.54±0.36)>prevent group(0.98±0.23)>normal group(0.35±0.31). Conclusions The expression of ICAM-1 were increased in the cerebral vessels of rats with Hhcy.The intervention of B Vitamins could decrease.
