不同品系小鼠急性胰腺炎并发肺损伤模型的比较

To Discuss the Influence of Different Variety of Mouse on the Model of Acute Pancreatitis association Lung Injury

摘要: 目的:比较小鼠的品系对诱导急性重症胰腺炎并发肺损伤模型的影响。方法:不同品系小鼠腹腔内注射雨蛙素诱导急性重症胰腺炎模型,雨蛙素(50ug/kg)稀释于生理盐水中,每小时注射1次,连续腹腔注射12h、20h;对照组腹腔注射等量生理盐水。各组末次注射后1h下腔静脉取血测血淀粉酶,取胰腺、肺脏组织行病理学检查。观察不同品系小鼠注射雨蛙素12h、20h后胰腺、肺组织形态学及血淀粉酶水平差异。结果:12h BALB/C小鼠急性胰腺炎并发肺损伤模型诱导成功:胰腺腺泡广泛变性、坏死及空泡化;炎症细胞大量浸润。肺部表现间质及部分肺泡腔有炎细胞浸润、肺不张、肺泡间隔增宽。12h昆明白实验组示血淀粉酶升高,胰腺腺泡广泛变性、酶原颗粒聚集腺泡细胞中,无炎症细胞浸润,无腺泡坏死及空泡化,肺组织未见异常。20h实验组BALB/C、昆明白小鼠造模成功,ICR小鼠仅出现胰腺轻微损伤。结论:Balb/C小鼠对药物反应灵敏,用药量少,节约时间与成本,更适用于急性重症胰腺炎模型的研究。

Abstract: To discuss the influence of different variety of mouse on the Model of Acute Pancreatitis association Lung Injury.Methods:The AP model was induced by injecting caerulein into the cavity of mice.Caerulein(50 ug/kg body weight)in 0.9% NaC1 at hourly intervals for 12h、20h;The control animals received the same volume of 0.9% NaCl without caerulein(0.2ml).The animals were killed after the last intraperitoneal injection.The severity was evaluated in terms of amylase level,histological changes.To observate the differences between KM, ICR and BALB / C at 12h, 20h.Results:BALB/C 12h experimental group with acute pancreatitis complicated with acute lung injury model succeed:pathology of pancreatic acinar appeared wide variability,and vacuolization acinar necrosis, a large number of infiltrating inflammatory cells.Interstitial and part of the alveolar space have more neutrophils, atelectasis, the gap widened alveolar. KM, ICR was not successful. 20h experimental group found that BALB / C, Kunming mouse model successful, ICR mice with acute pancreatitis only minor damage. Conclusion Balb / C mice of drug response, and less drug use, saving time and costs for acute necrotizing pancreatitis model.
