趋化因子CXCL12/CXCR4在非小细胞肺癌中的表达

Study on the Expression of Chemokine CXCL12/CXCR4 in Non-small Cell Lung Cancer

摘要: 目的:探讨趋化因子CXCL12及其受体CXCR4在非小细胞肺癌中的表达及其与临床病理特征间的关系。 方法:采用免疫组织化学SP三步法检测95例非小细胞肺癌(NSCLC组)患者及27例肺部良性肿瘤患者(对照组)的石蜡标本中CXCL12、CXCR4的表达情况,并比较其在不同临床病理参数患者间的表达情况。 结果:在NSCLC中,转移组的CXCL12和CXCR4的高表达率(67.2%、51.7%)均高于非转移组(35.1%、27.0%),差异均有统计学意义(P<0.05)。CXCL12、CXCR4的高表达率均随TNM分期的增高而逐渐增高,且III期的高表达率明显高于I期,组间卡方检验显示差异有统计学意义(P<0.05)。在对照组(非肿瘤组织)中,CXCL12和CXCR4的高表达率分别为0和7.4%(2/27),相应95例NSCLC的高表达率分别为54.7%(52/95)和42.1%(40/95),差异均有统计学意义(P<0.05)。CXCL12与CXCR4之间无明显相关性。CXCL12、CXCR4的高表达率均与入组患者的年龄、性别、肿瘤大小、组织学分型及分化程度之间差异无统计学意义(P>0.05)。 结论:CXCL12、CXCR4的表达与NSCLC的临床分期及淋巴结转移相关,其表达水平可作为判断NSCLC预后的参考指标。

Abstract: Objective: To investigate the relationship between the protein expression of chemokine CXCL12 and its receptor CXCR4 in tissues of non-small cell lung cancer(NSCLC ) and the clinical pathological features. Methods: Expression of CXCL12 and CXCR4 in NSCLC tissue(n =95) and benign tumor tissue(n=27) were detected by immunohistochemical staining. The relationship between CXCL12/CXCR4 protein expression and clinical pathologic features was also analyzed. Results: The high expression rates of CXCL12 and CXCR4 in NSCLC tissues with lymph node metastasis(67.2%、51.7%)were both significantly higher than those in NSCLC tissues without lymph node metastasis (35.1%、27.0%).Expression of CXCL12 and CXCR4 were closely associated with TNM stage(P<0.05),and the high expression rates in stage III were much higher than those in stage I. The high expression of CXCL12 and CXCR4 in control group(non-tumor tissues ) is much lower than that in NSCLC. There was no significant relationship between CXCL12 and CXCR4 expressions. We found no relationship between the positive rates of CXCL12 and CXCR4 and the age, gender, tumor size, histological type and degree of differentiation. Conclusion: Expression of CXCL12 and CXCR4 were associated with metastasis and TNM stage, and can be used to evaluate potential prognosis of the patients with lung cancer.
