三伏咳喘膏治疗支气管哮喘的实验研究

摘要: 摘要: 目的 观察三伏咳喘膏贴敷治疗对哮喘豚鼠白细胞介素-18(IL-18)、 嗜酸性粒细胞 (EOS) 及肺组织病理的影响, 并探讨其作用机制。方法 将 60 只豚鼠随机分为对照组、 模型组、 治疗 1 组(穴位贴药 1 h), 治疗 2 组(穴位贴药 3 h) ,治疗 3 组 (穴位贴药 6 h), 治疗 4 组 (注射地塞米松) ,每组 10 只。对照组腹腔注射生理盐水 1 mL, 其余组均腹腔注射 10%的鸡卵清白蛋白 1 mL 致敏, 饲养 14 d 后, 于第 15 天起将对照组之外的所有豚鼠置于 4 L 密闭玻璃罐中, 用 1%的鸡卵清白蛋白生理盐水对豚鼠雾化吸入, 30~60 s/次, 1 次/d, 连续 1 周。模型建立后第 2 天, 对照组及模型组于肺腧、 心腧、 膈腧处涂抹生理盐水, 纱布包扎固定; 治疗 1~3 组, 于同样穴位贴敷三伏咳喘膏(5 mm×5 mm×1 mm), 3 组分别贴治 1 h、 3 h、 6 h; 治疗 4 组于腹腔注射地塞米松磷酸钠注射液, 相应穴位处涂抹生理盐水固定;各组均是每天贴敷 1 次, 6 h/次, 共 7 d。结果 与模型组比较, 咳喘膏穴位贴敷各组血 IL-18、 EOS 水平明显下降, 病理学改变明显减轻, 穴位贴药 3 h 组效果最好。结论 咳喘膏穴位贴敷对哮喘豚鼠血 IL-18、 EOS 有明确影响, 可减轻哮喘豚鼠模型气道炎性损伤。

Abstract: Abstract: Objective To observe the effect of sanfukechuangao acupoint application on changes of interleukin- 18 (IL-18), eosinophils (EOS) and pulmonary pathology in asthma model of guinea pigs, and its underlying mechanism thereof. Methods A total of 60 guinea pigs were randomized into normal control group, asthma model group, acupoint-application 1 h group, acupoint-application 3 h group, acupoint-application 6 h group and dexamethasone group. Asthma model was es⁃ tablished by intraperitoneal injection of 10% ovalbumin (OVA, 1 mL) and forced inhalation of atomized 2% OVA. Skin prep⁃ aration and external application of Chinese medicinal herbs were made on back of guinea pigs, and "Feishu", "Xinshu", "Ge⁃ shu"were selected in acupuncture points). Levels of IL- 18 and EOS were detected by immunohistochemisty method. Re⁃ sults Compared with model group, the levels of IL-18 and EOS were significantly decreased, the pathological changes were significantly reduced in three acupoint- application groups. The effectiveness of acupoint-application 3 h group was the best in three groups. Conclusion Sanfukechuangao acupoint application can influence the levels of IL-18 and EOS effectively, and relieve phlegmonosis injuries in pulmonary tissue of asthma model of guinea pig.
