Heat Shock Preconditioning on Pancreatic Graft Reduces Posttransplant Ischemia Reperfusion Injury of Rat

Abstract: Abstract Objective:Rats were heat shock preconditioned to determine the mechanism that heat shock precondition(HPC) reduce ischemia reperfusion injury of rats model after pancreas transplantation. Method:Expression of heat shock protein70 (HSP70)of pancreas of rats who were heat shock preconditioned was detected at different time. rats model of ischemia reperfusion injury after pancreas transplantation were set up. Donator rats of high expression of HSP70 were regarded as experiment group for pancreas transplantation and donator rats of non- heat shock precondition as control group.Blood and pancreas of rats were examined after pancreas transplantation after 6 hours.Expression of HSP70 in pancreas was examined with western-blotting. Expression of TNF-α in pancreas was detected with immunohistochemistry. Hemodiastase of rats was determined with iodoamylum chromatometry.Pathological changes of pancreas was detected with microscope. Result:Expression of HSP70 of pancreas of donator rats achieves peak after HPC 12 hours, which has significant difference comparison with other time points (P<0.05), and which has significant difference comparison with no precondition group(P<0.01),recovery after HPC 48 hours. Levels of TNF-α、neutrophil and hemodiastase of pancreatic tissue significantly increased in control group comprared with that in sham group(P<0.01)。Levels of TNF-α、neutrophil and hemodiastase of pancreatic tissue significantly decreased in experiment group comprared with that in control group(P<0.05),and interstitial edema and hemorrhage of pancreas were reduced in experiment group. Conclusion:Heat shock preconditioning reduce ischemia reperfusion injury of rats model after pancreas transplantation. Mechanism of delaying protective effect after HPC correlate to production of HSP70.
