Effect of Silence of the PIM-1 Expression on Proliferation of Prostate Cancer Cell Line

Abstract: Abstract Objective: To investigate the effect of RNAi-mediated silencing of PIM-1 on the proliferation of prostate cancer cell. Methods: The recombinant plasmid vectors of RNAi were transfected into PC-3 cell line in vitro and the cell-counting assay were used to determine the proliferation activity of the stably-transfected cells. Flow cytometer(FCM) were used to detect the influence of the silence of PIM-1 on cell cycle and apoptosis in PC-3 cells. Results: Compared with the contrast groups, the ability of proliferation of the PC-3 cell line with silenced PIM-1 gene was suppressed remarkably. The percentage of the cells in G1 stage increased, while which in S stage decreased. At the same time, much more apoptosis cells were observed. Conclusion: Silence of PIM-1 gene can result in the block of the cell cycle, suppression of cell proliferation and increasing apoptosis in PC-3 cells. PIM-1 may serve as a effective target of gene therapy for prostate cancer in clinical practice .
