Abstract: The effect of Jinqi hypoglycemic table on expression of SREBP-1cmRNA in the liver of insulin-resistant rats SUN Nan,CUI Jing-qiu,GAO Zhi-hong, FENG Ping, The General Hospital Of Tianjin Medical University,Tianjin 300052,China 【Abstract】Objective To investigate the efficacy of Jinqi hypolgycemic tablet on expression of SREBP-1c mRNA in the liver of insulin-resistant rats. Methods Established a rat model of insulin resistance (IR) by feeding high-fat diet for 8 weeks, then randomly divided the insulin-resistant rats into two groups: HF group (n=9) that was fed with high-fat diet for another 6 weeks,,HF+JQ group (n=9) that was fed with high-fat diet and Jinqi hypolgycemic tablet for 6 weeks. Measured blood glucose, serum insulin, lipid concentrations and serum leptin, serum TNF-α. Semi-quantitative RT-PCR was used to detect the expression of SREBP-1c mRNA in the liver of rats at the end point. Results Compared with the HF group, serum insulin, OGTT 2h glucose, serum lipid, serum leptin and TNF-αconcentrations in HF+JQ group were decreased obviously, and insulin sensitive index(ISI)was higher. In addition, , the expression of SREBP-lc in the liver of HF+JQ group was significantly reduced compared with HF group[(0.75±0.06 vs 0.92±0.05),P<0.01]. Conclusion Jinqi hypolgycemic tablet can inhibit the expression of SREBP-1c mRNA in the liver of high-fat-fed rats and improve insulin resistance . 【Key words】 Insulin resistance, SREBP-1c, leptin, TNF-α, Hypoglycemic drugs (traditional Chinese medicine
